多因素复合刺激诱导建立大鼠气虚血瘀证动物模型的探索性研究

摘    要:目的:应用多因素复合刺激的方法建立大鼠气虚血瘀证动物模型.方法:选用SD大鼠,通过不可预知的慢性睡眠剥夺以及半高脂高糖饮食复合刺激,复制大鼠气虚血瘀证动物模型,检测血液流变学、血脂变化、血管活性分子改变以及血压和心室内压等客观指标方面变化.结果:通过对模型组动物的血液流变学、血脂变化、血管活性分子改变以及血压和心室内压客观指标方面的分析,发现模型大鼠在血瘀证的客观体征上有明显的改变,其中低切变率下的全血黏度明显升高(P<0.05),血浆AngⅡ和ET含量明显升高(P<0.05),血压和室内压有一定下降(P<0.05);与此相对应,经过补阳还五汤干预后前述指标出现明显改变,低切变率下的全血黏度和血浆AngⅡ明显降低(P<0.05),血压和室内压明显升高(P<0.05),表明该模型基本符合目前临床气虚血瘀证候客观变化特点.结论:采用不可预知的慢件睡眠剥夺以及半高脂高糖饮食复合刺激的方法可以成功建立气虚血瘀证大鼠动物模型,该模型的成功有望为将来血瘀证的进一步研究以及活血化瘀方药的筛选提供有益的实验手段.
