摘    要:目的:应用重组腺病毒AdmIL-12转染树突状细胞(DC)制备DC疫苗。方法:以重组腺病毒AdmIL-12转染DC后,用流式细胞仪检测DC的表型,用RT-PCR检测IL-12在DC内的表达,用EILSA法测定细胞培养上清液中的mIL-12和mIFN-γ的含量。通过IL-12刺激T细胞分泌IFN-γ的能力以检测DC分泌的IL-12的生物活性。结果:重组腺病毒AdmIL-12能高效转染DC,转染后DC的表型无显著改变,转染DC能大量分泌具有生物活性的IL-12。结论:腺病毒AdmIL-12成功转染DC,获得有效的DC疫苗。

Abstract:Objective:To transfect the Dendritic cells (DC) with recombinant adenovirus vector AdmIL-12, make vaccine of DC. Methods:DC was transfected by AdmIL-12 and its phenotype was measured with floweytometery. The expression of IL-12 in DC was valued with PT-PCR method. The value of IL-12 and IFN-γ in culture supernatants was nanlyzed by ELISA. The biological activity of IL-12 secreted from DC was tested by its ability to stimulate T cells to secret IFN-γ. Results: Recombinant adenovirus AdmIL-12 transfected DC effectively. After transfetion, the phenotype of DC had no significant changement,IL-12 was expressed more in DC, DC secreted more IL -12 with biological activity. Conclusion: AdmlL-12 transfeeted DC effectively. The vaccine of DC was made successfully.
