胃癌患者口腔菌群的分布特征

目的 利用高通量测序,研究胃癌患者口腔微生物的菌群分布特征,获得口腔菌群及牙周致病菌与胃癌的相关性的科学证据.方法 采集胃癌患者和健康对照人群口腔中的唾液及牙菌斑样本,PCR扩增,并利用Illumina Miseq测序平台,对各样本中16S rRNA V4区进行双端测序通过BIPES以及QIIME分析并比较不同人群口腔中牙菌斑和唾液样本中细菌群落结构及其多样性,聚类分析胃癌患者口腔菌群失衡的主要特征.结果 胃癌患者和健康人群的口腔菌群的交叠情况结果显示胃癌患者的口腔细菌更为复杂,菌群的组成和相对丰度分析发现,在胃癌组的唾液和牙菌斑样本中某些细菌的异常聚集,包括:红环菌目(Rhodocyclales)、梭菌目(Clostridiales)、肠杆菌目(Enterobacteriales)、假单胞菌目(Pseudomonadales)、暖绳菌目(Caldilineales)、菌目Synergistales.而芽孢杆菌目(Bacillales)、Streptophyta和立克次体目(Rickettsiales)3个在胃癌组中异常减少.结论 胃癌患者的口腔微生物群落结构与健康对照人群有所不同,提示口腔菌群发生相应改变与胃癌有一定的相关性.表明口腔微生物的检测有潜力作为未来胃癌早期诊断或筛查的一种手段.

Characteristics of oral microbiome in gastric cancer patients

Objective To investigate the characteristics of oral microbiome in gastric cancer patients, and to obtain scientific evidence of correlation between oral bacteria and periodontal pathogens and gastric cancer. Methods High-throughput sequencing was used to examine the total bacterial profile of saliva and plaque samples of 50 subjects,including 37 individuals with gastric cancer and 13 controls. The Venn diagram and species abundance cluster were generated from the data. Clustering analysis was used to obtain the main characteristics of the oral flora imbalance in patients with gastric cancer. Results The oral bacteria were more complex in the patients with gastric cancer. There was some abnormal aggregation of some bacteria in saliva and plaque samples of gastric cancer patients. Conclusion The structure of oral microbial community in patients with gastric cancer is different from that in healthy control population,suggesting that oral flora has some correlation with gastric cancer.
