三七总皂苷抗衰老的实验研究

摘    要:目的:从肝细胞线粒体DNA(mtDNA)、细胞因子的角度,探讨三七总皂苷(PNS)延缓衰老的可行性和可能的作用机制。方法:将实验大鼠随机分为青年对照组、衰老模型组、三七总皂苷组,每组12只。三七总皂苷组予三七总皂苷混悬液,余两组均喂普通饲料。检测实验肝细胞mtDNA的相对含量、血清SOD活性、MDA含量、血清白细胞介素-2(IL-2)、白细胞介素-6(IL-6)含量。结果:三七总皂苷能够降低衰老状态下大鼠肝细胞mtDNA相对含量及血清MDA含量,提高血清SOD活性,与衰老模型组比较,有显著性差异(P〈0.05);同时还能够增加衰老状态下大鼠血清IL-2含量,降低IL-6含量,与衰老模型组比较,有显著性差异(P〈0.05)。结论:三七总皂苷具有减轻D-半乳糖诱导亚急性衰老大鼠模型的氧化损伤,保护肝细胞mtDNA,提高机体的免疫功能作用。

Abstract:Objective :To study the anti-aging feasibility and possible mechanism of Panax Notoginseng saponins (PNS) from the aspect of hepatocyte mitoehondrial DNA(mtDNA) and eytokines. Methods:The rats were randomly divided into young eontrol group,aging model group and PNS group,each group with 12. MtDNA relative content in rat hepatocyte was analyzed by SDS alkaline denaturation and serum SOD activity was estimated by xanthine oxidase method. MDA content was measured by thiobarbitude acid eolouration. The serum interleukin-2(IL-2) and interleukin-6(IL-6) levels were determined by ELISA method. Results:PNS reduced mtDNA relative content,serum MDA content and serum level of IL-6 in all aging rat hepatocyte significantly compared with the aging model group(P〈0.05). PNS promoted serum SOD activity and increased serum level of IL-2 compared with the aging model group (P〈0.05). Conclusion:PNS could relieve oxidative damage of aging rats induced by D-galaetose,protect hepatoeyte mtDNA and enhance immune function.
