Dermcidin作为新的肺癌生物标志物的研究

目的研究Dermcidin(DCD)作为新的肺癌生物标志物的可行性。方法收集病理确诊肺癌的患者手术前血清、肺癌组织和癌旁组织共31例,用ELISA方法对肺癌患者和健康对照组血清中DCD浓度作比较;利用定量聚合酶链反应(quantitative polymerase chain reaction,q PCR)方法对同一患者肺癌标本和癌旁标本进行ΔCt值的比较;利用Western blot方法对同一患者肺癌标本和癌旁标本DCD的表达量进行比较;同时利用免疫组化方法对同一患者肺癌组织和癌旁组织DCD的表达程度进行比较。结果 ELISA结果发现肺癌血清中DCD浓度高于健康对照组,差异有统计学意义(P0.05);q PCR结果显示肺癌组与癌旁组ΔCt值的差异有统计学意义(P0.05);Western blot结果进一步表明肺癌组织中DCD的表达量高于癌旁组织,差异有统计学意义(P0.05);免疫组化结果也发现肺癌组织中DCD表达水平高于癌旁组织,差异有统计学意义(P0.05)。结论 DCD与肺癌密切相关,可能成为肺癌潜在的分子标志物。

New tumor marker dermcidin for lung cancer

Objective To investigate the feasibility of dermcidin(DCD) as a new biomarker for lung cancer.Methods Thirty-one serum before operation,lung cancer tissues and adjacent tissues of the patients who were pathological diagnosed as lung cancer were collected.The concentration of DCD were compared by ELISA between lung cancer group and healthy control group,△Ct value of DCD were compared by qPCR between lung cancer group and adjacent tissues group,the expression of DCD were compared by Western blot between lung cancer group and adjacent tissues group.The expression level of DCD were compared by immunohistochemical methods between lung cancer group and adjacent tissues group.Results ELISA results showed that the serum level of DCD was significantly higher in lung cancer group than the healthy controls (P＜ 0.05).qPCR showed that there was significant difference in △Ct value between lung cancer tissues and adjacent tissues (P＜ 0.05);Western blot further showed that expression of DCD in lung cancer tissues was significantly higher than that of adjacent tissues (P＜0.05);Immunohistochemistry also found that expression level of DCD in lung cancer tissues was significantly higher than that of adjacent tissues (P＜0.05).Conclusion It suggested that DCD was closely related to lung cancer,it may be a potential molecular marker for lung cancer.
