肝素抗涎腺腺样囊性癌细胞裸鼠肺血管粘附研究

摘    要:目的 通过观察肝素对人涎腺腺样囊性癌肺高转移细胞株 (ACC -M )细胞在裸鼠肺、肝组织的粘附影响 ,证实其对腺样囊性癌肺高转移细胞肺粘附的抑制作用。方法 裸鼠腹腔注射肝素后 ,经尾静脉注入氚标胸腺嘧啶核苷 (3 HTdR)标记的ACC -M细胞 (3 H -ACC -M ) ;分别检测细胞注射后 2h、6h、18h肺、肝组织单位重量的每分钟同位素脉冲数 (CPM )。结果 单位重量肺组织每分钟放射性计数CPM值 :空白对照组 2h、6h、18h分别为 1875 .75± 11.5 3、164 4.5 0± 2 7.2 0和 14 66.5 0± 12 5 .95 ;2 0 0单位肝素组 2h、6h、18h分别为 912 .0 0± 118.5 5、918.0 0± 44 .0 6和 766.60± 77.66。同一时间肝素组肺组织CPM值明显低于对照组 ,有显著性差异 (P <0 .0 0 1)。单位重量肝组织CPM值在空白对照组和肝素组的相同时间点无显著性差异 (P >0 .0 5 )。结论 肝素腹腔注射后可明显降低肺组织内ACC -M细胞对血管内皮细胞粘附数量 ,但对相同时间肝组织内ACC -M细胞粘附数量无显著影响 ,提示肝素具有抑制腺样囊性癌肺转移的作用

Abstract:Objective To investigate the effect of intraperitoneal administration of heparin on the lung-metastasis character of highly lung metastatic adenoid cystic carcinoma cell strain (ACC-M).Methods 3HTdR labeled ACC-M cells were injected by intravenous infusion after intraperitoneal injection with 200 units heparin.4 mice of each group were killed at 2h,6h and 18h after infusion, respectively. The relative radioactivity (CPM) in lung and liver was detected. Result 3H-activity per gram in lung of heparin group was lower than in control group. There was significant difference between two groups (P<0.001). There was no difference between two groups in liver (P>0.05).Conclusion The results of this study suggest that the highly lung metastasis characteristic of ACC-M may be partially inhibited by the intraperitoneal use of heparin.
