眼镜蛇神经毒的分离、纯化及其镇痛作用

摘    要:中华眼镜蛇粗毒经二次CM-Sephadex C25柱层析分离、纯化获得眼镜蛇神经毒。聚丙烯酰胺圆盘电泳显示一条区带。在小鼠热板模型及大鼠电尾嘶叫模型,眼镜蛇神经毒显示有镇痛作用,并有剂量-效应关系。

Abstract:The neurotoxin(NT) was fractionated and purified from the cobra venom by chro-motography on CM-sephadex C25. The purified NT obtained showed only one zone in the gel of disc electrophoresis. The purified fraction(NT) completely blocked the transmission in neuromuscular junction of chick biventer cervicis muscle and did not markedly affect the contraction of frog heart. It was demonstrated that the antinociceptive action caused by NT showed a good dose-response relationship in two models, mouse hot plate and vocalization induced by electrical stimulation of the rat's tail. The paw-lick latencies of the mice of both NT groups in different dose (0.023mg/kg, 0.03rog/kg) were significantly prolonged than that of the control group. In the rats, the vocalization threshold of NT group rose significantly than that of the control group too. The analgesic potency of NT with a dose of 0,022mg/kg was approximently equal to that of morphine with a dose of 4 mg/ kg.
