大鼠血浆中牡荆素鼠李糖苷的质量浓度测定

目的建立测定大鼠血浆中牡荆素鼠李糖苷质量浓度的反相高效液相色谱（RP-HPLC）法。方法以四氢呋喃-乙腈-0.5%冰醋酸（20：2.5：77.5）为流动相,血浆样品经简单的甲醇沉淀蛋白后,采用RP-HPLC法测定。结果血浆中牡荆素鼠李糖苷质量浓度的线性范围为0.2～80.0μg/mL,平均相对回收率为89.27%～98.25%,日内和日间精密度的RSD均小于10.79%,最低检测限为50ng/mL。结论建立的RP-HPLC法简便快速、灵敏度高,适用于牡荆素鼠李糖苷的药代动力学研究。

Determination of Vitexin-Rhamnoside in Rat Plasma

Objective To establish a RP-HPLC method to detect the concentration of vitexin-rhamnoside in rat plasma.Methods The mobile phase consisted of tetrahydrofuran-acetonitrile-0.5% glacial acetic acid（20：2.5：77.5,v /v /v） was used.After pretreated by methanol,the plasma samples were analyzed by RP-HPLC.Results The linear range of vitexin-rhamnoside concentration was 0.2-80.0 μg /mL.And the recovery rate was 89.27%-98.25%.Both the within-day and between-day RSD were less than 10.79%.The minimum detective limit was 50 ng /mL.Conclusion This RP-HPLC method is accurate,stable and sensitive,which can be applied to the pharmacokinetic study of vitexin-rhamnoside.
