| 摘 要： | 目的 :探讨梗阻性黄疸形成中肿瘤坏死因子 -α(TNF -α)与肝细胞凋亡的变化。方法 :72只Wistar大鼠随机分为正常组、假手术组和胆总管结扎手术组 ,以结扎胆总管 (ligatingcommonbileduct ,LCD)的Wistar大鼠为模型 ,采用放射免疫法 ,测定血清中TNF -α含量 ;TUNEL法检测大鼠肝细胞凋亡状态。结果 :LCD术后 3、7、14dTNF -α含量明显高于正常组和假手术组 ,并于LCD术后 7～ 14d达到高峰。结论 :Wistar大鼠在梗阻性黄疸形成中TNF -α明显增高诱导肝细胞凋亡 ,对肝功能损害起着一定作用。

| Abstract: | Objective To investigate the changes of tumor necrosis factor (TNF-α) and hepatocellular apoptosis in obstructive jaudice rats.Methods 72 wistar rats were divided into three groups: normal, sham ligating common bile duct and ligating common bile duct. The amount of specium TNF-α was decteted by radioimmunity. Apoptosis was detected by terminal-deoxynucleotidyl transferase mediated dUTP nick labeling (TUNEL). Results The amount of specium TNF-α in LCD group was higher than that in normal and sham LCD groups at 3?7?14?21 days after LCD.TNF-α reached its peak from 7 to 14 days post LCD.Conclusion TNF-α and hepatocellular apoptosis are increased obviously in obstructive jaudice rats and play an important role in the injury liver. |
