双苯氟嗪对拟痴呆大鼠和老龄大鼠的脑保护作用研究

摘    要:目的观察双苯氟嗪对拟痴呆大鼠和老龄大鼠的脑保护作用。方法选用雄性SD大鼠,按体煎随机分为正常对照组、模型对照组、尼莫地平5.0mg/kg刚性对照药组、双苯氟嗪3.0mg/kg组和双举氟嗪10.0mg/kg组。除对照组大鼠腹腔内注射生理盐水外,其他各组均腹腔注射氯化铝致痴呆模型,并分别给予生理盐水、双萃氟嗪或尼莫地平。测定血清中超氧化歧化酶(SOD)活性和丙二醛(MDA)的含量,并观测脑细胞胆碱乙酰转移酶(ChAT)的表达量;另选用21月龄的老龄大鼠,随机分为模型对照组、尼莫地平5.0mg/kg组和双苯氟嗪5.0mg/kg组,同时选用3月龄成年雄性大鼠作为正常对照组。观察灌胃给予双苯氟嗪对神经细胞内游离钙离子浓度(Ca^2+)及细胞凋亡百分率的影响。结果双苯氟嗪可以显著升高拟痴呆大鼠血清中SOD、降低血清中MDA及增加海马和皮层两脑区ChAT的表达量,并且降低老龄大鼠海马和皮层两脑区细胞(Ca^2+)。及细胞凋亡百分率。结论双苯氟嗪对痴呆大鼠具有脑保护作用,其作用机制可能与提高机体抗氧化能力、增强胆碱能神经功能、抑制细胞钙担载及细胞凋亡有关。

Abstract:Objective To investigate the neuroprotective effects of dipfluzine on dementia-mimetic rats and aged rats.Methods Dementia rats model induced by intraperitoneal injection of AlCl_3 were given dipfluzine to determine serum superoxide dismutase(SOD) activity and malondialdehyde(MDA) content and to observe expressions of choline acetyl transferase(ChAT).The aging rats aged 21 months selected were treated with dipfluzine by oral administration to observe(Ca~(2+))i and cellular apoptosis percentage.Results Dipfluzine significantly increased SOD activities and the expression of ChAT in the neurons,decreased serum MDA content and(Ca~(2+))i and cellular apoptosis percentage.Conclusions Dipfluzine has neuroprotective effects on dementia rats,which may be correlated with its inhibitory effects on peroxidation and calcium overload in the neurons and the effect of enhancing the function of cholinergic neurons and inhabiting apoptosis in the neurons.
