基质金属蛋白酶与肺癌的侵袭和转移

摘    要:目的 :探讨基质金属蛋白酶 (m atrix m etalloproteinases,MMPs)、端粒酶与肺癌侵袭和转移的关系。方法 :运用酶谱法 (zym ography) ,TRAP(端粒末端重复扩增 )法定量分析肺癌和癌旁组织 MMP- 2、MMP- 9、端粒酶活性与肺癌侵袭和转移的关系。结果 :MMP- 9几乎见于所有肺癌组织 ,且活性较强 ,MMP- 2表达较弱 ,仅部分表现为阳性。癌组织平均 MMP- 9含量高于癌旁 ,但差异无显著性 (P=0 .5 98)。 2 7例肺癌中有 12例癌组织 MMP- 9含量高于癌旁 ,另外 15例低于癌旁 ,分别占 4 4 .4 %和 5 5 .6 % ,癌组织和癌旁组织 MMP- 9含量差异有显著性 (P=0 .0 4 5 )。结核瘤 MMP- 9含量明显高于癌旁 (P=0 .0 12 ) ,平均含量高于癌组织 ,但差异无显著性。不同组织类型的肺癌 MMP- 9活性不同 (P<0 .0 5 ) ,细支气管肺泡腺癌比腺癌和鳞癌 MMP- 9活性低。尚无证据表明 ,肺癌端粒酶与 MMP- 9活性、淋巴结转移显著相关。结论 :MMP-9不仅在预测肺癌病人预后、术后随访、判断复发等方面有重要意义 ,并且 ,随着各种 MMPs抑制剂研究的深入 ,以 MMPs为靶的抗肺癌转移和血管生成的抗癌策略有望进入临床应用阶段。在以 MMP- 9判断肺癌侵袭转移能力时 ,要注意与结核瘤鉴别

Abstract:Objective:To investigate the relations hip among matrix metalloproteinases (MMPs),telomerase and metastasis of lung cancer. Methods:The activities of MMP-2 and MMP-9 of lung cancer and adjacent tissues were measured by zymography.Telomerase activity was detected using telomeric repeat amplification protocal enzyme-linked immunoadsorbent assay(TRAP-ELISA).Results:MMP-9 was observed in almost all samples as a proenzyme and expressed strongly,while MMP-2 only expressed weakly in some tumor tissue.The average MMP-9 activity of tumor tissues are higher than that of adjacent tissues,but there is no significant difference(P=0.598).15(55.6%) percent of 27 tumor tissues' MMP-9 is lower than adjacent tissues'.MMP-9 in tuberculoma is significantly higher than tumor adjacent tissues(P=0.045).MMP-9 activities are varied with histological types.The activities of MMP-9 in Alveolar carcinoma were much lower than that in other non-small cell lung cancer(NSCLC).Conclusion:MMP-9 are important factors affecting lymph node metastasis of lung cancer and are hopeful target of inhibit NSCLC invasion and metastasis .But we should diagnosis it with tuberculoma.
