用cDNA表达阵列分析遗传性癫癎易感大鼠大脑皮质的基因表达

摘    要:应用cDNA阵列技术寻找遗传性癫差异表达基因 ,为从基因水平探讨癫的发病机理打下基础。采用AtlasTM RatcDNAExpressionArray建立遗传性癫易感性P77PMC大鼠和正常Wistar大鼠大脑皮质基因表达谱 ,用图像分析仪分析两者基因表达谱差异。结果发现有 15个差异表达基因 ,其中 13个基因在P77PMC大鼠大脑皮质中高表达而在正常Wistar大鼠大脑皮质中低表达 ,2个基因在正常Wistar大鼠大脑皮质中高表达而在P77PMC大鼠大脑皮质中低表达。结果提示 ,P77PMC大鼠与正常Wistar大鼠大脑皮质存在多个差异表达基因 ,这些差异表达的基因可能在癫的发生、发展中扮演了重要角色。

Abstract:This study aimed to search the differential expression genes of genetical epilepsy and to establish the groundwork of exploring the pathogenesis of epilepsy on gene level by using cDNA array technology. First, gene expression profiles of the cerebral cortex of genetical epilepsy prone P77PMC rats and normal Wistar rats were established by using Atlas TM Rat cDNA Expression Array. Then the difference of gene expression profiles was analyzed with image analysis instrument, and 15 differential expression genes were discovered. Among them, 13 genes were high expressed in the P77PMC rats but low expressed in the normal Wistar rats, 2 genes were high expressed in the Wistar rats but low expressed in the P77PMC rats. In conclusion, there are several differential expression genes between P77PMC rat and Wistar rat. All these genes may play important roles in the pathogenesis of epilepsy.
