冠心丹参片对大鼠心肌缺血再灌注损伤的保护作用

 目的观察冠心丹参片对健康成年SD大鼠心肌缺血再灌注损伤的保护作用。方法SD大鼠以结扎冠脉30min再通90min造成心肌缺血再灌注损伤模型,通过观察血流动力学、生物化学和组织学指标,评价大鼠缺血再灌注前冠心丹参片预处理对左心室收缩压(LVSP)、左心室发展压(LVDP′)、左心室舒张末压(LVEDP)、左心室等容期压力最大变化速率(±dp/dt_max)等指标的影响;通过测定再灌注90min后血中乳酸脱氢酶(LDH)、肌酸激酶(CK)活性和心肌组织病理学观察,对药物保护心肌作用进行评价。结果大鼠冠心丹参片100,200mg·kg^-1,ig可有效改善缺血再灌注心脏的缩舒功能;降低缺血再灌注末血中LDH和CK活性;组织病理学检查表明:与溶剂对照组比较,冠心丹参片预处理组能明显减轻心肌组织的损伤。结论冠心丹参片有抗大鼠缺血再灌注引起的心肌损伤作用。

Effects of Guanxin Danshen Tablet on Myocardium Injury Induced by Ischemia-Reperfusion in Anesthetized Rats

OBJECTIVE To observe the cardioprotective effects on Guanxin Danshen tablet on myocardium injury induced by ischemia-reperfusion in anesthetized adult SD rats.METHODS The myocardial injury of rats induced by ischemia-reperfusion was produced by the ligation of left coronary artery for 30 min followed by 90 min reperfusion. The influence of Guanxin Danshen tablet on the changes of biochemistry,histology and hemodynamics, such as LVSP, LVDP′,LVEDP and dp/dt_max,were evaluated. The serum lactate dehydrogenease(LDH) and creatine kinase(CK) activities were assayed by the end of the reperfusion. RESULTS The systolic and diastolic function were improved,and the serum LDH and CK activities were reduced in Guanxin Danshen tablet 100 and 200 mg·kg^-1 ig group.The histology changes were manifested. The pretreatment of Guanxin Danshen tablet alleviated the myocardial damage distinctly, compared with ischemia-reperfusion group.CONCLUSION The experimental results demonstrate the effects of Guanxin Danshen tablet on myocardial injury induced by ischemia-reperfusion in anesthetized rats.
