摘    要:目的建立HPLC法测定乐脉胶囊中芍药苷和丹酚酸B的含量。方法芍药苷和丹酚酸B均采用C18(4.6mm×250mm,5μm)色谱柱;流动相分别为乙腈-0.05%磷酸(13:87)和乙腈-甲醇-1.7%甲酸溶液(4:27:69);检测波长分别为230和286nm,柱温为30℃,流速为1.0ml·min^-1。结果芍药苷在0.20403—4.0806μg范围内得芍药苷回归方程Y=1×10^6X+2631(r=0.9999);丹酚酸B在0.12573~2.5147μg范围内得回归方程Y=937308X+560947(r=0.9996),两组分分别与峰面积呈良好的线性关系。芍药苷的平均回收率为98.9%,RSD为1.8%;丹酚酸B平均回收率为100.4%,RSD为2.8%。结论该方法结果准确,重复性好,可作为乐脉胶囊质量控制的定量方法。

Abstract:Objective To establish an HPLC method for the determination of paeoniflorin and salvianolic acid B in Lemai capsules. Methods Chromatographic experiments on paeoniflorin were performed on Cls column, the mobile phase consisted of acetonitrile-0.05% phosphoric acid (13: 87) ,the detection wavelength was 230 nm,the column temper-ature was 30 ℃ ,and the flow rate was 1.0 ml ·min^-1. Similarly,chromatographic experiments on salvianolic acid B were performed on C18 column (4. 6 mm × 250 mm, 5 μm), the mobile phase was aeetonitrile-methanol-1.7% formic acid (4:27:69) ,the detection wavelength was 286 nm, the column temperature was 30 ℃, and the flow rate was 1.0 ml·min^ - 1. Results The linear equations were Y = 1 × 10^6 X + 2631 ( r = 0. 9999) for paeoniflorin and Y = 937 308 X + 560 947 ( r = 0. 9996) for salvianolic acid B, respectively. The linear range was 0.204 03 - 4.0806 μg for paeoniflorin and 0. 201 10 - 2. 5147 μg for salvianolie acid B, respectively. The average recovery was 98.9% ( RSD = 1.8% ) for paeoniflorin and 100.4% ( RSD = 2.8% ) for salvianolic acid B, respectively. Conclusion This method is simple and accurate with good repeatability. It can be used for quantity control of Lemai capsules.
