枇杷花水提物防治小鼠急性酒精性肝损伤研究

摘    要:目的初步观察枇杷花水提物(AELF)对急性酒精染毒小鼠的防治效果。方法取雄性昆明种小鼠,随机分为空白组、模型组、联苯双酯阳性药组、不同剂量AELF(400、800、1600mg·kg^-1)组,每组10只。按0.10ml/10g剂量灌胃给药10d,末次给药2h后一次性灌胃给予50%(Ⅳy)无水乙醇溶液0.12ml/10g,禁食不禁水16h。测定外周血象,血清中ALT、AST、ADH活性及肝匀浆中MDA、TG含量。结果与模型组相比,AELF各剂量组能明显降低WBC、AST、血清及肝脏TG水平,均以中剂量效果最好(P〈0.01,P〈0.01,P〈0.01,P〈0.05);高剂量组能够明显降低血清ALT及肝脏MDA水平(P〈0.01,P〈0.05)。结论AELF可以通过降低染毒小鼠转氨酶和TG水平,提高机体抗氧化能力,对小鼠急性酒精性肝损伤起到一定的预防性保护作用。

Abstract:Objective To investigate the effect of the aqueous extract of loquat flower (AELF) on acute alcoholic liver injury in mice. Methods The mice were randomly divided into 6 groups:control group, model group, positive drug group (Bifendate) ,and AELF groups at different doses (400,800,1600 mg·kg^-1). Each group has 10 mice. Drugs were given once a day for 10 consecutive days,0.10 ml/10 g a time. 50% ( V/V ) dehydrated alco-hol of 0.12 ml/10 g was fed orally 2 h after the last administration except in control group. Then the animals were fasted for 16 h with water drinking allowed. Results Compared with model group, the AELF group at the dose of 800 mg ·kg^-1 underwent a marked decrease in the level of aspartate amino transferase (AST), triglyceride (TG) of the serum and liver( P 〈0.01, P 〈0.01, P 〈0.05) and the amount of WBC returned to normal( P 〈0.01), while in the 1600 mg·kg^-1 group the level of alanine amino transferase (ALT) in serum and the concentration of malondialdehyde (MDA) in liver homogenates was reduced significantly ( P 〈 0.01, P 〈 0.05 ). Conclusion AELF can lower the level of ALT, AST, TG and improve the anti-oxidation ability of organisms. It has proactively protective effect on acute alcohol-induced liver injury in mice.
