摘    要:目的对引入界面增强剂磺化聚醚醚酮的新型PEEK/HA生物材料进行生物相容性评价,研究该材料对骨环境细胞的氧化应激作用。方法选用成骨细胞MG-63、成纤维细胞3T3作为研究对象,比较材料对两种细胞产生ROS和MDA水平的影响,并观察两种细胞在HA质量占比30%的PEEK/SPEEK/HA材料上的生长形貌。结果在PEEK和SPEEK材料上培养导致成骨细胞MG-63和成纤维细胞3T3氧化应激水平上升,SPEEK材料相比PEEK材料能降低细胞氧化应激水平,PEEK/HA材料对成骨细胞MG-63和成纤维细胞3T3氧化应激作用相对PEEK和SPEEK材料减弱,与PEEK/HA生物材料相比,PEEK/SPEEK/HA生物材料使两种细胞氧化应激水平更低,当HA含量达到30%,成骨细胞MG-63氧化应激水平接近正常,并且两种细胞在该材料表面生长正常不受到限制。结论HA与SPEEK引入能降低细胞氧化作用。

Abstract:Objective To evaluate the biocompatibility of new PEEK/HA biomaterials with sulfonated polyether ether ketone as interface reinforcer and study the oxidative stress of PEEK/SPEEK/HA biomaterial on bone environment cells. Methods Osteoblasts MG-63 cells and fibroblasts 3T3 cells were selected as the research objects to compare the effects of biomaterials on ROS and MDA levels of the two kinds of cells and to observe the growth morphology of the two kinds of cells on PEEK/SPEEK/30% HA materials. Results The oxidative stress level of MG-63 cells and 3T3 cells increased when cultured on PEEK and SPEEK materials, but PEEK/HA was lower than PEEK and SPEEK materials. Compared with PEEK materials, SPEEK materials can reduce the oxidative stress level of cells, PEEK/SPEEK/HA biomaterials made the oxidative stress level of MG-63 cells and 3T3 cells lower than PEEK/HA biomaterials, when the HA content reached 30%, the oxidative stress level of MG-63 cells was close to normal and the growth of the two cells on the surface of the biomaterials was unrestricted. Conclusion The introduction of HA and SPEEK could reduce the oxidative effect of cells.
