电针对慢性应激抑郁模型大鼠行为学及海马BDNF表达的影响

摘    要:目的 观察电针对慢性应激抑郁模型大鼠行为学及海马脑源性神经营养因子(BDNF)表达的影响,初步探讨电针治疗抑郁的机制.方法 将SD大鼠40只分为正常组、模型组、电针组、氟西汀组,除正常组不造模外,其它3组采用慢性轻度不可预见性应激配合孤养复制抑郁模型.通过敞箱试验、糖水消耗量观察抑郁模型大鼠行为学改变情况,并运用免疫组化法检测电针对抑郁模型大鼠海马神经元细胞BDNF表达的影响.结果 抑郁模型大鼠敞箱试验中水平穿越格数、竖立次数、理毛时间、24h糖水消耗量均明显减少,体重增加缓慢,中央格停留时间明显增加,与正常组比较差异均有统计学意义(P<0.01);而电针组及氟西汀组大鼠的水平穿越格数、竖立次数、理毛时间、24h糖水消耗量、体重均有明显增加,中央格停留时间明显减少,与模型组比较差异亦均有统计学意义((P<0.01或0.05).模型组BDNF免疫反应阳性神经元数目明显减少,平均灰度值较正常组明显增加(P<0.05或0.01);而电针组和氟西汀组大鼠海马BDNF免疫反应阳性神经元数目明显增加,平均灰度值明显减小,与模型组比较差异亦有统计学意义(P<0.05或0.01).结论 电针能明显改善抑郁大鼠模型的各项行为学指标,增加海马BDNF的表达,与氟西汀同样具有抗抑郁作用.

Abstract:Objective To observe the effects of electro-acupuncture on behavior and the level of brain-derived neurotrophic factor(BDNF) in the hippocampus of rats with chronic stress depression. Methods Forty Sprague-Dawley male rats were randomly divided into four groups:a normal control group, a model control group, an electro-acupuncture group and a fluoxetine hydrochloride control group. All rats except the control groups were singly test and by the volume of sugar solution drunk in 24 hours. The levels of BDNF in the CA1 and CA3 pyramidal and dentate gyrus of the hippocampus were investigated immunohistochemically. Results Compared with the normal control group, the model rats'crossing squares, rearing, and grooming in the open field test and the volume of sugarsolution drunk were all less, while their time in the central square was clearly greater. Fluoxetine or electro-acupuncture could significantly improve behavior. Compared with the model group, BDNF immunoreactivity in the hippocampal neurons of the electro-acupuncture group was increased significantly and the average gray value of neurons decreased.Conclusions Electro-acupuncture can improve behavior and the level of BDNF in the hippocampal neurons of depressed rats. It has a significant antidepressant effect.
