中药复方双术油抗肿瘤机制的研究

目的本研究旨在探讨复方双术油(BELY)的体内外抗肿瘤作用机制。方法采用CCK-8试剂盒检测对K562、MDA-MB-231、LOVO和HepG2的影响;建立小鼠S180及H22肿瘤模型,观察BELY对该模型的作用;Elisa法测定IL-2、IL-6、IL-12及TNF-α的水平,探讨BELY对荷瘤小鼠免疫功能的影响;流式细胞仪测定荷瘤小鼠外周血CD4+/CD8+。结果 BELY对HepG2、K562、Lovo、MDA231均有一定的生长抑制作用,并有良好的量效关系。BELY显著抑制S180和H22的生长,提高荷瘤小鼠的胸腺指数、脾指数和体重。BELY可明显升高小鼠血清中IL-2、IL-6、IL-12和TNF-α的水平,外周血CD4+/CD8+明显升高。与生理盐水组比较,P<0.01或P<0.05。结论 BELY既可以抑制肿瘤细胞生长,又可通过增强免疫从而达到双相抗肿瘤的作用。

Anti-tumor Mechanism of Chinese Herbal Compound Shuang-zhu Oil

Obeject To investigate the anti-tumor mechanism of BELY in vitro and in vivo anti-tumor effect.Method through the use of CCK-8kit to detect K562 and MDA-MB-231,the LOVO and HepG2 cell proliferation;through murine sarcoma S180 and of H22 transplant tumor model to observe the effection of BELY on tumor-bearing mice;Use Elisa to detect of cytokines IL-2,IL-6,IL-12 and TNF-alpha's level to reflect the effect of BELY on immune function of tumor-bearing mice;measured by flow cytometry of tumor-bearing mice peripheral blood T cell subsets of CD4 + and CD8 +,and calculate the ratio of the two types of cells.Results BELY had a certain degree of inhibition on HepG2 and K562,Lovo,MDA231 cells and had the dose-effect relationship.BELY significantly inhibited the growth of the murine sarcoma S180 and of H22,and improve the thymus index,spleen index and weight of the tumor-bearing mice,BELY could be significantly higher serum CD4 + / CD8 + compared with the blank control group(P <0.01 or P <0.05).Conclusion BELY not only directly inhibit tumor cells,but also enhancing immune to achieve the dual proposition of inhibitl tumor.
