人参皂苷Re对大鼠心室肌细胞钠、钾离子通道的影响

摘    要:目的:观察人参皂苷Re对大鼠心室肌细胞电压依赖性的钠、钾离子通道的影响,揭示其抗心律失常的作用机制。方法:采用酶分法获得单个大鼠心室肌细胞,在全细胞膜片钳技术模式下,记录电压依赖性钠通道、瞬时外向钾通道和内向整流钾通道电流,并观察不同浓度的人参皂苷Re对电流幅度的影响。结果:给予人参皂苷Re(10,100μmol·L-1)后钠通道电流幅度呈现减小的趋势,对通道电流幅度的抑制率分别为(25.5±5.7)%(n=3)和(46.2±18)%(n=4)。给予人参皂苷Re(10,100μmol·L-1)对瞬时外向钾电流幅度略有抑制,洗脱后电流恢复,其中100μmol·L-1人参皂苷对电流的抑制率为(13±9)%(n=3)。给予人参皂苷Re(10,100μmol·L-1)后内向整流钾电流幅度呈现逐渐减小的趋势,药物洗脱后电流完全恢复,(10,100μmol·L-1)对通道电流幅度的抑制率分别为(16.3±8.7)%(n=5)和(35.4±11.2)%(n=5)。结论:人参皂苷Re能够抑制心室肌细胞电压依赖性的钠通道、瞬时外向钾通道和内向整流钾通道电流发挥抗心律失常的作用。

Abstract:Objective: To investigate the effects of Ginsenoside Re on voltage-dependent sodium current and potassium current in rat ventricular myocytes,and to investigate the mechanism of antiarrhythmia. Methods: The effect of Ginsenoside Re on sodium current( I Na),transient outward potassium current( Ito) and inwardly rectifie- d potassium current( I k1) were assessed by applying whole-cell patch clamp technique in single cardiac myocytes. Results: Ginsenoside Re( 10,100μmol·L-1)) was shown to inhibit the altitude of I Na and the inhibition ratio was 25.5 ± 5.7%( n = 3) and 46.2 ± 18%( n = 4). It was shown a little inhibition by Ginsenoside Re( 100μmol·L-1)) and its effects were reversible completely after washout. The inhibition ratio of Ito was 13 ±9%( n =3) in 100μmol·L-1).Ginsenoside Re( 10,100μmol·L-1)) inhibited the altitude of I k1and the inhibition ratio was 16. 3 ± 8. 7%( n = 5) and 35. 4 ± 11. 2%( n =5). Conclusion: Ginsenoside Re can inhibit sodium current and potassium currents to produce antiarrhythmic effect.
