摘    要:目的:探讨研究预培养环节对分支杆菌噬菌体生物扩增法试验结果的影响。方法:对166例临床诊断为肺结核患者的标本,经液化处理后均进行过夜组(预培养24h)及不过夜组(室温放置6h后)分支杆菌噬菌体生物扩增试验,试验方法按结核分支杆菌噬菌体裂解试剂盒(FAST PlaqueTBTM)操作手册操作,对照分析二者的阳性率和污染率。结果:166例肺结核患者标本,过夜组分支杆菌噬菌体试验结果阳性率为59.0%;非过夜组分支杆菌噬菌体试验结果阳性率为57.8%,二者的阳性率接近,差异无统计学意义(x2=0.496,P>0.05),过夜组污染率为13.9%,明显高于未过夜组的7.2%,二者的差异有统计学意义(x2=3.86,P<0.05)。结论:分支杆菌噬菌体生物扩增法试验标本处理后室温放置6h比预培养1d效果好,可以提前一天报告结果,建议临床推广应用。

Abstract:Objective: To probe into prep-cultivating time's effect on phage expansion experiment of tubercle bacillus.Method: The example made from 166 case of lung patients were liquefied,then separated into overnight(prep-cultivating for 24 hours) group and non-overnight group(keeping 6 hours at room temperature) to perform the bacillus phage expansion experiment,by means of a method based on FAST Plaque TBTM operation manual.A comparative analysis has been done on positive reaction rate and negative reaction rate between such two groups.Result: For such 166 case of lunger exemplar,overnight group has a 59.0% positive reaction rate based on the result of bacillus phage experiment,non-overnight group has a 57.8% positive reaction rate,and such rate is so close that the difference is no meaning for statistics(x2=0.496,P>0.05).However,pollution rate for overnight group is 13.9% which is higher apparently than 7.2% for non-overnight group,and such difference has a meaning for statistics(x2=3.86,P<0.05).Conclusion: For bacillus phage experiment exemplar after being prep-treated,it is better to keep them for 6 hours at room temperature than prep-cultivating for one day;the former has a good effect and can get the result one day in advance.It is our suggestion to promote such practice in clinic.
