染料木黄酮对前列腺癌细胞系PC-3生物学行为影响的意义

摘    要:目的观察染料木黄酮(genistein)对前列腺癌细胞 PC-3生物学行为的影响 ,探讨 genistein预防前列腺癌的可能性.方法用 MTT法绘制生长曲线,观察 0, 10,20, 40 μ mol/L的 Genistein对 PC-3细胞生长能力的影响; PC-3细胞经 Genistein处理 3 d后,流式细胞术观察 Genistein对前列腺癌细胞 PC-3细胞周期的影响, Transwell小室法重建基底膜侵袭模型研究 Genistein处理后细胞侵袭性的改变.结果 经 Genistein处理后, PC-3细胞的生长受到抑制,作用 3 d时的药物半数致死量(IC50)为 25 μ mol/L;细胞周期改变主要为 G2/M阻滞,凋亡率(AI)明显增加, 0 ,10,20, 40 μ mol/L组的 G2/M的百分比和 AI分别为 14.9%, 27.4% ,33.1% ,31.9%和 0, 6.5% ,14.2% ,25.4%.侵袭能力分别下降到未加药组的 31.8%, 8.6%和 3.96%.结论 Genistein通过引起 PC-3细胞 G2/M阻滞,诱导 PC-3细胞凋亡,从而抑制 PC-3细胞的恶性增殖以及降低 PC-3细胞的侵袭能力可能是 Genistein降低前列腺癌的发病率的一个重要原因.Genistein有可能成为预防前列腺癌的药物之一.

Abstract:AIM:To investigate the effect of genistein on the biological behavior of PC-3 cell line of carcinoma of prostate and explore the possibility of genistein used as a preventive drug. METHODS:PC-3 cells were exposed to conditional media containing 0, 10,20, 40 μ mol/L genistein for three days . The proliferation of PC-3 cells was studied by MTT analysis.The effects of genistein on cell cycle were detected by FCM. We also investigated the abilities of the genistein-treated cells to invade through the basement membrane incursive model reconstructed by using Transwell chambers. RESULTS:The proliferation was inhibited in a time-dependent and a dose-dependent manner after treating with genistein,and the IC50 of genisein was 25 μ mol/L after 3 days treatment. Cell cycle was blocked at G2/M stage,the proportion of G1 stage decreased in a time-dependent and a dose-dependent manner,and the percent of G2/M cells was 14.9%, 27.4% ,33.1% ,31.9% in the 0,10,20 and 40 μ mol/L genistein group respectively.The percent of apoptosis cells was 0%, 6.5% ,14.2% ,25.4% respectively. The ability of genistein-treated cells to invade the reconstituted basement membrane decreased to 31.8%, 8.6% and 3.96% in comparison with that of the control group. CONCLUSION:Genitein inhbits the invasion of PC-3 cells in a dose-dependent manner. Genistein may act as a protective drug for prostatic cancer by inhibiting the proliferation,inducing apoptosis and decreasing the invasive ability of PC-3 cells.
