摘    要:目的研究兔急性肺栓塞前后血浆血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)及一氧化氮(NO)表达水平的变化,以及应用尿激酶(UK)和氯沙坦(Losartan,LOS)对其的影响,为急性肺栓塞的治疗探讨一种新方法。方法健康中国大耳白兔40只,建立急性肺栓塞模型,随机分为栓塞组、UK组、LOS组和联合组,每组10只。四组分别于栓塞前、栓塞后用药前、用药后1h、2h、4h抽静脉血,分别测血浆中AngⅡ和NO的含量。结果 AngⅡ的表达量在栓塞组和LOS组随时间延长逐渐升高,LOS组较栓塞组低,在UK组和联合组造模后即开始升高,2h达高峰,此后开始下降;NO在所有组中1h时达最低点,此后栓塞组仍处于较低水平,各治疗组则呈上升趋势,各组间及组内差异均有统计学意义(P0.05)。结论尿激酶联合氯沙坦可显著降低急性肺栓塞兔血浆AngⅡ的表达,并升高NO表达,及时降低肺栓塞引起的肺动脉高压并保护内皮功能,为急性肺栓塞的治疗提供一种新方法。

Therapeutical Effect of UK Combined with Losartan on Acute Pulmonary Embolism in Rabbits

Abstract:Objective To observe the changes of Ang Ⅱ and NO before and after acute puhnonary embolism in rabbits and the influence of UK and LOS on the two in an effort to find a new method to treat acute pulmonary embolism. Methods Forty healthy rabbits were used to establish models of acute pulmonary embolism and divided into four groups at random (ten rabbits per group):Group Embolism, Group UK, Group LOS, Group UK and LOS. The venous blood from each group was drawn to detect the levels of Aug Ⅱ and NO before embolism and at Oh ,1h, 2h, and 4h after embolism. Results Ang Ⅱ level rose gradually in Group Embolism and LOS, and began to rise in Groups Uk and Combination after embolism and achieved a peak 2h after embolism, and then began to decline. The NO level was the lowest in all groups at lh , then lh lowered in Group embolism but began to rise in other groups, with a significant difference between groups and between intra-groups (P〈0.05). Conclusion UK combined with LOS can decline the Aug Ⅱ level and rise the NO level in acute pulmonary embolism in rabbits obviously and lower pulmonary artery hypertension induced by pulmonary embolism and protect the blood vessel endothelium, providing a new method to treat acute pulmonary embolism.
