苄基丹皮酚肟抗血栓活性的初步研究

摘    要:目的探讨苄基丹皮酚肟(benzyl paeonol oxime,BPO)的抗血栓活性。方法采用全自动凝血因子分析仪,检测家兔血浆PT、APTT和TT;比浊法检测血小板聚集率;ELISA法检测大鼠血浆中PC、PS、AT-Ⅲ、GPⅡb/Ⅲa和cAMP含量。结果BPO对APTT有延迟作用,对ADP、AA、胶原诱导的血小板聚集有抑制作用,可升高血浆中蛋白C的浓度,降低抗凝血酶Ⅲ的浓度,但未引起蛋白S的浓度变化;同时BPO可升高血浆中GPⅡb/Ⅲa的浓度,而对cAMP的浓度无影响。结论BPO具有优良的抗凝血活性和抗血小板聚集活性,其抗凝血机制可能与增强PC活性和促进AT-Ⅲ与凝血酶结合有关;抗血小板聚集机制可能与抑制纤维蛋白原和GPⅡb/Ⅲa的结合有关。

Abstract:Objective To study the antithrombotic activity and antiplatelet aggregation of benzyl paeonol oxime(BPO).Methods The automatic coagulation factor analyzer is used to detect the prothrombin time,activated partial thrombin time and thrombin time with the plasma of rabbits.The platelet aggregation are measured by light transmission aggregometry.The concentrations of plasma protein C,protein S,antithrombinⅢ,GPⅡb/Ⅲa and cAMP in plasma of rats are detected by ELISA.Results BPO prolonged the APTT.BPO significantly inhibited platelet aggregation induced by ADP,AA and collagen.BPO increased the concentration of PC and reduced the concentration of AT-Ⅲ,but did not change the concentration of PS.BPO also increased the concentration of GPⅡb/Ⅲa in plasma,but it had no obvious effect on the concentration of cAMP.Conclusion BPO has excellent anticoagulant activity and antiplatelet aggregation activity,its anticoagulant mechanism may be related to enhance PC activity and promote the AT-Ⅲcombined with thrombin.The mechanism of antiplatelet aggregation may be related to inhibiting the binding of fibrinogen and GPⅡb/Ⅲa.
