醛固酮合成酶基因多态性及生活习惯与急性脑梗死易感性的关系

摘    要:目的研究醛固酮合成酶(CYP11B2)基因-344T/C多态性及烟酒茶嗜好相互作用与急性脑梗死(ACI)易感性的关系。方法采用病例对照研究方法,调查研究对象的生活习惯,采用聚合酶链反应限制性内切酶长度多态性(PCR-RFLP)方法检测CYP11B2的基因型。结果病例组CYP11B2基因TC和CC基因型的频率娃著高于对照组,分别为38.12%和9.97%;相对于TT基因型,暴露于TC和CC基因型人群的OR值分别为1.72和1.88。病例组C等位基因的频率也显著高于对照组,为29.03%;相对于T等位基因,C等位基因的OR值为1.57。此外研究还显示:CYP11B2基因TC或CC基因型与吸烟相互作用可增加ACI的易感性,而该基因型与饮酒、饮茶无显著相互作用。结论当个体携带TC或CC基因型时,ACI的易感性增加,该两种基因型与吸烟相互作用,可增加ACI的易感性。

Abstract:Objective To investigate the relationship between aldosterone synthase (CYP11B2) gene-344T/C polymorphism, dietary habits and the risk of acute cerebral infarction (ACI).Methods The dietary habits data were collected by case-control study.-344T/C polymorphism of CYP11B2 gene was monitored by polymerize chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results The frequency of TC, CC in ACI cases were significantly higher than those in controls (38.12%, 9.97%). The odds ratios (OR) for those exposed to TC and CC genotypes were 1.72 and 1.88, comparing with that to TT genotype. The frequency of C allele in ACI cases was 29.03%, significantly higher than that of controls. The OR for those exposed to C allele was 1.57, comparing with that to T allele. In addition, TC or CC genetype interacted with smoking to increase the risk of ACI; No interaction was observed between TC, CC genetype and alcohol drinking or tea intake.Conclusions The CYP11B2-344 TC or CC genetype might increase the risk of ACI development and the genetype might also interact with smoking to increase the susceptibility of ACI.
