凉膈散对内毒素致大鼠急性肺损伤T辅助细胞漂移的影响

摘    要:目的 探讨凉膈散对内毒素致大鼠肺损伤时机体致炎、抗炎反应的分子机制.方法 用内毒素脂多糖(LPS)复制大鼠急性肺损伤模型.分别用高、中、低剂量的凉膈散水煎剂灌胃给药,并与地塞米松做对照,于不同时间点检测外周血及支气管肺泡灌洗液(BALF)中T辅助细胞1(Th1)和T辅助细胞2(Th2)的含量,观察Th1/Th2比值的变化.结果 模型组大鼠外周血与BALF中Th1/Th2比值逐渐降低,在4、8、16h时间点与空白组比较差异有统计学意义(P<0.01),中、高剂量组与地塞米松组的Th1/Th2比值在8h和16h与模型组同时间点比较有所提高,差异有统计学意义(P<0.05或P<0.01).结论 凉膈散在一定程度上纠正了Th1/Th2细胞比例失衡,这可能是其对急性肺损伤具有保护作用的机制之一.

Abstract:Objective To investigate the molecular mechanism of inflamation development and anti-inflamation response of Lianggesan Decoction(Decoction for cooling the chest)on endotoxin-induced acute lung injury rats.Methods The rat models with acute lung injury were made by lipopolysaccharide(LPS)through intravenous injection.The high,middle and low dosage of Lianggesan Decoction were given by gastric perfusion and compared with dexamethasone(DXM).The contents of Th1 and Th2 in peripheral blood and bronchoalveolar lavage fluid(BALF)were measured at different time points to observe the changes in Th1/Th2.Results In the model group,Th1/Th2 in peripheral blood and BALF was gradually lowering,the difference was significant compared with those of the blank group(P<0.01)at 4h,8h,16h.In the middle and high dosage groups and DXM group,Th1/Th2 was getting higher at 8h and 16h compared with that of the model group with a significant difference(P<0.05 or P<0.01).Conclusion Lianggesan Decoction corrects the Th1/Th2 imbalance,which might be one of the mechanisms in protecting the acute lung injury.
