雌二醇预处理对大鼠脑缺血再灌注损伤脑组织HSP70表达的影响

摘    要:目的:观察雌二醇对脑缺血再灌注损伤脑组织HSP70表达的影响。方法:大鼠分假手术对照组、脑缺血再灌注组和雌二醇治疗组,每组32只,采用线栓法建立大鼠局灶性脑缺血再灌注损伤模型,每组分别于再灌注3h、6h、12h、24h处死8只动物,于视交叉前后2mm脑冠状切开,取闭塞侧组织制备连续性切片.免疫组化技术检测脑组织中HSP70的表达。结果:3组脑血管内皮均有HSP70的表达。假手术组皮层和纹状体无HSP70表达:雌二醇治疗组与脑缺血再灌注组再灌注后各时间点皮层、纹状体均有HSP70的表达,且雌二醇治疗组脑缺血再灌注后12h、24h皮层HSP70表达高于脑缺血再灌注组。结论:雌二醇预处理,可以诱导大鼠脑缺血再灌注脑组织HSP70的表达,对缺血性脑损伤可能有一定的保护作用。

Abstract:Aim: To investigate the effects of estradiol on HSP70 expression in rat brain tissue after cerebral ischemia GAAB2 reperfusion injury. Methods: Rat cerebral ischemia GAAB2 reperfusion injury model was established with the middle cerebral artery occlusion (MCAO) by reversible inserting a nylon thread method. A total of 72 male Wistar rats were allocated into MCAO group,sham operation group,and estradiol treatment group,32 rats in each group.At 3 h,6 h,12 h and 24 h after reperfusion,8 rats were executed,respectively.Serial sections for immunohistochemistry were used to detect the expression of HSP70. Results: HSP70 was expressed in the vascular endothelial cells in all the groups. In sham operation group HSP70 was absent in cerebral cortex and corpus striatum.HSP70 expression was detected in the cortex and corpus striatum of estradiol treatment group and MCAO group at different time points,and the expression in estradio treatment group was higher than that of MCAO group at 12 h and 24 h. Conclusion: Pretreatment with estradiol can increase HSP70 expression in cerebral focal ischemia-reperfusion injury tissue,and may protect brain from ischemic injury.
