摘    要: 目的:建立以葡萄糖为参比测定多糖或寡糖含量的方法。方法:对苯酚-硫酸法的测定条件进行了改进,测定了几种单糖对葡萄糖的吸光度校正系数k′i,并用它们计算寡糖或多糖的含量。结果:对4种寡糖和4种多糖(包括均聚糖和异聚糖)的含量用改进的方法进行了测定,该方法的相对误差小于4%,具有良好的重复性,线性范围含糖量0~30μg。结论:利用在改进的测定条件下测得的各种单糖的校正系数,可以以葡萄糖为标准品对组成己知的寡糖或多糖的含量进行测定。

Abstract:OBJECTIVE:To establish a method for determination of the content of oligo and polysaccharides,using only glucose as the standard.METHOD:The absorbance correction coefficents of several monosaccharides were determined with the modified phenol-sulfuric acid method and then used in the calculation of the content of saccharides.RESULTS:The modified method was used for determination of the saccharide content of four oligosaccharide and four polysacchride samples,and was shown to be excellent in accuracy and reproducibility,with relative error less than 4%.The linear range of saccharide content was 0~30 μg.CONCLUSION:The correction coefficients determined with the modified phenol-sulfuric acid can be successfully used for determination of the content of oligo-and polysaccharides with only glucose as the standard.
