摘    要:目的 为寻求一种更安全有效的腹膜透析液,本实验观测了多种渗透性溶质对培养的人腹膜间皮细胞增殖的影响。方法 将人腹膜间皮细胞体外培养,细胞增殖率以^3H-胸腺嘧啶脱氧核苷摄取量表示,细胞膜损伤通过测定培养上清液中乳酸脱氢酶的含量反映。渗透性溶质包括晶体溶质(葡萄糖、棉子糖、山梨醇、蔗糖、甘露三糖、甘油、必需氨基酸混合物)和胶体溶质(葡聚糖-70、麦芽糊捕、羟乙基淀粉-20)。结果 所有的渗透性溶质都以浓度(或渗透压)依赖方式抑制细胞的增殖。晶体溶质的抑制作用大于胶体溶质。大部分高浓度的晶体溶质对细胞膜有损伤作用,但高浓度的胶体溶质对细胞的损伤并不明显。结论 多种渗透性溶质对腹膜间皮细胞的增殖均有不同程度的毒性影响,胶体溶质毒性作用小。体外腹膜间皮细胞培养可为探索新的腹膜透析液提供一种筛选方法。

Abstract:Objective To evaluate the acute effects of various solutions at high osmotic concentration on the proliferation of human peritoneal mesothelial cells (HPMC) in vitro. Methods HPMC proliferation was assessed by using 3?H-thymidine incorporation. The damage to HPMC membrane was measured by the augmented release of cytosolic enzyme lactic dehydrogenase (LDH). The tested osmotic solutions included 50 mmol/L or 100 mmol/L crystalloid agents (glucose, raffinose, sorbitol, sucrose, mannitol, glycerol, a mixture of essential amino acids) and 3.5% or 7% colloid agents (dextran 70, maltodextrin, hydroxyethylstarch 20). Results All of the solutions at high osmotic concentration and neutral pH inhibited the growth of HPMC in a concentration-dependent way. The inhibition by the crystalloid agents was stronger than that by the colloid agents. At higher concentration (100 mmol/L), most of the crystalloid also damaged the HPMC membrane, but no significant injury to HPMC was observed caused by the colloid agents at higher concentration (7%). Conclusion The results suggest that the toxic effect of osmotic solutes on the proliferation of HPMC depends on the hyperosmolarity. The colloid solutes seem to be less toxic to mesothelial cell proliferation.
