寒凝证类痛经大鼠模型建立的探讨

摘    要:目的:探讨寒凝证类痛经大鼠模型的建立。方法:将处于动情问期的3月龄SD雌性大鼠60只,按照随机数字法分为6组,每组10只,分别为:空白对照组、盐水对照组、类痛经模型组、冷冻5d组、冷冻5d+给药组、冷冻10d+给药组。空白大鼠同步抓取不做任何处理,类痛经模型组、冷冻5d组、冷冻5d+给药组、冷冻10d+给药组均每日给予大鼠皮下注射苯甲酸雌二醇注射液,连续10d,第1d及第10d日注射0.5mg/只,第2~9d每日0.2nag/只。末次给药1h后,类痛经模型组、冷冻5d+给药组、冷冻10d+给药组腹腔注射缩宫素2U/只。盐水组每日给予同等剂量的生理盐水。每天在注射以后,冷冻5d组、冷冻5d+给药组、冷冻10d+给药组放入超低温冰柜冷冻4h,在2h时打开冰柜换气5s,冰柜温度维持在~25℃。观察每分钟扭体评分、寒证表征评分、全血还原黏度、红细胞聚集指数以及红细胞刚性指数。结果:与空白对照组比较,冷冻5d组、冷冻5d+给药组、冷冻10d+给药组寒证表征评分明显升高(P〈0.01);类痛经模型组、冷冻5d组和冷冻5d+给药组的全血还原黏度高切均明显升高(P〈0.05),冷冻5d+给药组的红细胞刚性指数明显升高(P〈0.05):与盐水组比较,类痛经模型组、冷冻5d+给药组、冷冻10d+给药组的每分钟扭体评分明显升高(P〈0.05)。结论:全身冷冻法可造成大鼠寒凝证;通过寒凝证模型结合类痛经模型的制备方法,成功建立了寒凝证类痛经大鼠模型,该模型符合中医证候模型的造模思路及特点。

Discussion on Establishment of Dysmenorrhea Rat Model with Cold Stagnation Syndrome

Abstract:Objective : To explore the establishment of dysmenorrhea rat model with cold stagnation syndrome. Method : 60 anestrous 3 -month -old female SD rats were divided into six groups according to the random number, n = 10, inclu- ding blank control group, saline control group, dysmenorrhea model group, freeze 5 d group, freeze 5 d + injection group, and freeze 10 d + injection group. Blank rats were synchronically crawled without any treatment. Dysmenorrhea model group, freeze 5 d group , freeze 5 d + injection group, freeze 10 d + injection group were given daily subcutane- ous injection of estradiol benzoate injection for 10 consecutive days, on the first day and lOth day injected 0. 5 mg for each time, during day 2 to day 9, injected 0.2 mg everyday. 1 h after last injection, rats were intraperitoneally injected oxytocin 2 U for dysmenorrhea model group,freeze 5 d + injection group and freeze 10 d + injection group. Saline group were given the same dose of saline. After the injection, the freeze 5 d group, freeze 5 d + injection group and freeze 10 d + injection group were put into the ultra - low temperature freezers for 4 hours, and the freezer ventilation was open at 2 hours for 5 seconds and the freezer was maintained at a temperature of - 25 ℃. The writhing score per minute, cold syndrome characterization score, whole blood reduced viscosity, and erythrocyte aggregation index and erythrocyte rigidity index were observed. Result : Compared with the blank control group, cold syndrome characterization scores of freeze 5 d group, freeze 5 d + injection group and freeze 10 d + injection group were significantly higher (P 〈 0.01 ) ; full blood re- duced viscosity high shear of dysmenorrhea model group, freeze 5d group and freeze 5 d + injection group were signifi- cantly increased (P 〈 0.05), erythrocyte rigidity index of freeze 5 d + injection group was significantly higher ( P 〈 0.05) ; compared with the saline group, writhing score per minute of dysmenorrhea model group, freeze 5 d + injection group and freezel0 d + injection group was significantly higher (P 〈 0.05 ). Conclusion: Systemic freezing can cause cold stagnation syndrome in rats. To combine preparation of dysmenorrhea model with cold stagnation model,we success- fully established the dysmenorrhea rat model with cold stagnation syndrome.
