金威消石颗粒对大鼠血液学影响的实验研究

摘    要:目的:研究由中医验方研制而成的金威消石颗粒对大鼠血常规及凝血功能的影响。方法:健康SD大鼠40只,雌雄各半,随机选入金威消石颗粒高、中、低剂量纽及空白对照组,分别经胃灌注不同浓度的金威消石颗粒混悬液和蒸馏水,12.5mL/kg,每日2次,每周给药6d,连续3个月。然后经腹腔动脉取血1mL,检查血常规和凝血酶原时间。结果:金威消石颗粒中、低剂量组大鼠红细胞压积(Hct)偏高、血红蛋白(Hb)偏低,低剂量组红细胞(RBC)偏高,中剂量组淋巴细胞(Lymph)%偏低和嗜酸性粒细胞(Eos)%偏高,与空白对照组相比均有显著性差异(P〈0.05),但均在正常范围内。各剂量组大鼠PT与空白对照组相比无显著性差异(P〉0.05)。结论:金威消石颗粒给药3个月对大鼠血常规及凝血功能未产生明显药物相关性不良反应。这对金威消石颗粒临床应用提供可靠的理论依据。

Abstract:Objective : To study the impact of Jinwei Lithiasis - eliminating Granule, which was made according to the emperical TCM recipe, on the routine blood test and coagulation function in rats. Methods: Forty healthy SD rats including 20 male and 20 female, were randomly distributed to three pilot groups ( high - , middle - and low - dose groups) and blank control group. Each group had 5 male and 5 female rats. Jinwei Lithiasis - eliminating Granules (40.0 g/L,27.0 g/L, 13.0 g/L) and distilled water were perfused by gavage for each groups' rats, respectively, twice a day,6 days in a week ,lasting for 3 months. The perfusion dosage was 12.5 mL/kg for each rat. After anaethesia by intraceliacal injection of 25% urethane (0.4 mL/100 g) ,the abdominal cavity of each rat was opened and 1 mL blood sample was taken from each rat' s celiac artery for blood routine test and prothrombin time measurement. Results : As compared with blank con- trol group, higher hematocrit level and lower hemoglobin level (F = 3. 922 and 3.817, P 〈 0.05 ) in the middle and low dosage groups, and higher red blood cell count (F = 3. 392 ,P 〈 0.05) in low dosage group, as well as lower lymphocyte percent ( F = 3. 652, P 〈 0.05 ) and higher eosinophilic granulocyte percent ( F = 3. 51, P 〈 0.05 ) in middle dosage group were found,whereas all of them were within normal ranges. There were no significant differences in prothrombin time test between three Jinwei Lithiasis -eliminating Granules groups and blank control group (F = 0. 330, P 〉 0.05 ). Conclusions : Jinwei Lithiasis - eliminating Granules exhibited no drug - related adverse reaction on the routine blood test function and coagulation function in rats after 3 months' medication. It could offer a reliable theoretical basis for the clin- ical application of Jinwei Lithiasis- eliminating Granules.
