狼毒乙素和新狼毒素A的抗肿瘤作用

摘    要:目的:观察狼毒乙素(ECB)对 U14宫颈癌的荷瘤小鼠,以及新狼毒素A对雄激素非依赖性前列腺癌(AIPCa)裸鼠移植瘤的抑制作用。方法:① U14宫颈癌的腋下移植瘤小鼠各50只,各随机分成5组,实验组灌胃分别给予狼毒乙素(ECB)高、中、低组(12、10、8mg/kg),阳性药组灌胃给予环磷酰胺(50mg/kg),模型组给予等量生理盐水,给药14d后处死动物剥离瘤组织称质量,计算抑瘤率。②雄性裸小鼠20只,皮下接种DU-145细胞,随机分成用药组和对照组。于癌细胞种植第2天起,对照组每天喂饲0.9%氯化钠溶液,用药组每天喂饲新狼毒素A15rag·kg-1·d-1。记录用药第2—7周末时移植瘤的平均体积。结果:①狼毒乙素(ECB)高、中、低剂量组对小鼠 U14宫颈癌的抑瘤率分别为66,40%,49,61%,44.14%;②用药组裸小鼠移植瘤的生长明显受到抑制,第3周末至第7周末的移植瘤体积均显著小于对照组(P值均〈0.001)。第7周结束时与对照组比较,用药组的生长抑制率达到65.34%。结论:狼毒乙素(ECB)对小鼠宫颈癌U14有显著抑制作用;新狼毒素A可以显著抑制DU-145肿瘤的生长,提示该药对AIPCa有抗癌活性。

Abstract:Objective :To research the inhibition effect of ECB on U14 cervical cancer and NCA on androgen independ- ent prostate cancer(AIPCa) xenografts in nude mice. Methods:l. Fifty cervical cancer bearing mice were randomly divid- ed into five groups, which received ECB high - dose, middle - dose and low - dose ( 12 mg/kg, 10 mg/kg, 8 mg/kg) , cyclophosphamide (50 mg/kg) and saline respectively for 14 days. The tumor tissues were cut off,weighed and the inhi- bition rate was calculated. 2. DU - 145 cells were subcutaneously injected to 20 male BALB/c - nude mice and those mice were randomly divided into administration group and control group. From day 2 after the injection, the mice were fed with 0. 9% saline solution in the control group and NCA( 15 mg. kg- 1 .' d- 1 )in the NCA group every day. The aver- age size of xenografts was measured every week from week 2to week 7. At the end of week 7, all mice were executed and the tumors were removed. Results:l. The inhibition rate of U14 cervical cancer was 66.40% for high - dose ECB( P 〈 0. 01 ) ,49.61% for middle - dose ECB ( P 〈 0.01 ) , 44.14 % for secondary low ECB H Dripping Pills ( P 〈 0.01 ) . 2. The growth of xenografts in the NCA group was significantly inhibited, and the tumors were significantly smaller than those in the control group from the end of week 3 to week 7 ( all P 〈 0. 001 ). Compared with control group, the growth inhibiting rate of the tumors in the NCA group was 65.34% at the end of week 7. Conclusion.ECB can significantly in- hibit the growth of U14 cervical cancer bearing mouse;NCA can obviously inhibit the growth of DU- 145 cells,indicating that NCA has the anti -tumor activity to AIPCa.
