高原红细胞增多症骨髓红系祖细胞凋亡异常的研究

摘    要:目的:了解高原红细胞增多症(HAPC)骨髓红系祖细胞在细胞凋亡方面是否存在异常;方法:应用造血干细胞体外培养技术对红系祖细胞进行动态观察,培养72小时以后细胞染色观察细胞形态,应用凋亡细胞检测盒了解细胞凋亡发生率;结果:高原红细胞组与正常对照组均发生细胞凋亡,但发生率程度不一,高原红细胞凋亡少于正常对照(P<0.05);结论:高原红细胞增多症红系祖细胞存在凋亡异常,需进一步研究是否存在基因表达异常。

Abstract:Objective:To investigate whether the apoptosis in high altitude polycythemia (HAPC) is different from that in normal.Methods:The bone marrow cells were cultured to observe the erythroid progenitor cells in different time points.Trypan blue exclusion was used to determine the viability. Apoptosis was assessed by cell morphology and PCD-ASSAY-KIT.Results:At different time points,the number of apoptosis cells in normal condition is smaller obviously than that in HAPC (P<0.05).Conclusions:It is suggested that there is different apoptosis in erythroid progenitor cells by comparing the HAPC with the normal.
