绞股蓝提取物对糖尿病肾病大鼠血液流变学和炎性因子影响的实验研究

摘    要:目的初步探讨绞股蓝提取物对糖尿病肾病大鼠血液流变学和炎性因子的影响。方法将40只Wistar大鼠随机分为空白对照组、模型组、绞股蓝大剂量组(5mg/kg)和小剂量组(2.5mg/kg);模型组、大剂量组和小剂量组大鼠使用腹腔注射链脲佐菌素方法制作糖尿病肾病模型;绞股蓝大小剂量组按照上述剂量给予绞股蓝提取物灌胃,1次/d,持续10周,空白对照组和模型组则给予同剂量蒸馏水灌胃;实验结束时腹主动脉取血检测血液黏度、总胆固醇、三酰甘油及炎症因子。结果绞股蓝提取物可以有效降低糖尿病肾病大鼠血液黏度、血浆黏度及总胆固醇和三酰甘油水平,与模型组比较差异具有统计学意义(P〈0.05);同时可明显降低糖尿病肾病大鼠血清TNF-α、IL-6和CRP水平,与模型组比较差异具有统计学意义(P〈0.05)。结论绞股蓝提取物具有改善糖尿病肾病大鼠血液流变学及微炎症状态的作用,值得进一步研究。

Abstract:Objective To investigate the effects of Gynostemma extract on the hemorheology and inflammation factors of diabetic nephropathy rats. Methods Forty wistar rats were randomly divided into control group, model group, high-dose of Gynostemma group (5 mg/kg) and low-dose of Gynostemma group (2.5 mg/kg); streptozotocin injection were given to the rats in model group, high-dose Gynostemma group and low-dose Gynostemma group by intraperitoneal as the diabetic nephropathy models. The rats in high and low dose of Gynostemma groups were given the above dose of Gynostemma extract by intragastric administration, 1 time/d, for 10 weeks; The rats in blank control group and model group were treated with the same dose of distilled water. The viscosity, total cholesterol, triglycerides and inflammatory factors were detected by extracting the abdominal aortic blood of all rats at end of the experiment. Results The blood level of viscosity, plasma viscosity, total cholesterol and triglyceride in rats with diabetic nephropathy were effectively reduced by Gynostemma extract, comparing with model group, and the difference had statistical significance (P 0.05); Meanwhile, the blood level of TNF-α, IL-6 and CRP were either lowered by Gynostemma extract, compared with model group, the difference had statistical significance (P 0.05). Conclusion Gynostemma extract can effectively improve the hemorheology and micro-inflammatory state of the rats with diabetic nephropathy, which deserves further study.
