温阳活血方对慢性马兜铃酸肾病大鼠肾损害的干预作用

摘    要:目的:观察温阳活血方对慢性马兜铃酸肾病肾损害的干预作用。方法:将48只雄性SD大鼠随机分为5组:(1)正常对照组(n=8):予生理盐水灌胃;(2)模型组(n=10):按关木通水煎液10ml·kg^-1·d^-1(相当于关木通40g·kg^-1·d^-1,马兜铃酸A2.6mg·kg^-1·d^-1)给大鼠灌胃;(3)中药组(n=10):在模型组基础上,再予温阳活血方30g·kg^-1·d^-1灌胃;(4)西药组(n=10):在模型组基础上,再予科素亚33.3mg·kg^-1·d^-1灌胃;(5)中西药结合组(n=10):在模型组基础上,再予温阳活血方+科素亚灌胃。20周末,收集大鼠尿液测定24h尿蛋白、NAG、β2-MG,腹主动脉取血用于测定Scr、BUN、RBC、Hb。结果:与正常对照组比较,模型组大鼠24h尿蛋白、NAG、β2-MG、Scr、BUN均明显升高(P〈0.01,P〈0.05),而血Hb、RBC均明显下降(P〈0.01);与模型组比较,治疗组大鼠24h尿蛋白、NAG、β2-MG、Scr、BUN均明显下降(P〈0.01,P〈0.05),而血RBC、Hb均明显升高(P〈0.01,P〈0.05)。结论:温阳活血方对慢性马兜铃酸肾病大鼠肾损害有一定的保护作用,能降低尿蛋白和尿NAG、β2-MG的排泄,改善肾功能和贫血。

Abstract:Objective:To study the effects of Wen Yang Huo Xue Fang(WYHXF) on urine protein, renal function, Hb and RBC in rats with chronic aristolochic acid nephropathy. Methods: Fourty - eight male SD rats were randomly divided into five groups. C-roup A( n = 8) were treated with normal .saline( 10 ml·kg^-1·d^-1 ). Group B( n = 10) were treated with caulis Aristolochia Manshuriensis decoction(Aristolochia Manshuriensis 40 g·kg^-1·d^-1, Aristolochic acid A2.6 mg·kg^-1·d^-1 ) to establishe the model. Group C( n= 10) were treated with WYHXF(30 g·kg^-1·d^-1 ) after the rats model established with the above-mentioned method. Group D(n =10) were treated with Losartan Potassium Tablets (33.3 mg·kg^-1·d^-1) after the rat model established with the above - mentioned method. Group E( n - 10) were treated with WYHXF( 30 g·kg^-1·d^-1 ) and Losartan Potassium Tablets ( 33.3 mg·kg^-1·d^-1) after the rat model established with the above - mentioned method. After 20 weeks, rats' urine was collected for the detection of 24 hour urine protein, NAG and β2 - MG. Blood from abdominal aortic artery was sampled for the detection of Scr, BUN, RBC and Hb. Results:Compared with those in the group A,the levels of 24 - hour urine protein, NAG,β2 - MG,Scr and BUN were significantly increased in the group B (P〈 0.01, P〈 0.05), while the level of RBC and Hb were significantly decreased in the group B( P〈 0.01 ). Compared with those in the group B, the levels of 24 - hour urine protein, NAG, β2 - MG, Scr and BUN were significantly decreased( P 〈 0.01, P 〈 0.05 ), while the levels of RBC and Hb were significantly increased in the group C, D and E (P 〈 0.01, P 〈 0.05). Conclusion: WYHXF could reduce the levels of urine protein, NAG and β2 - MG and improve renal function and alleviate anemia in chronic aristolochie acid nephropathy.
