赖氨酸6-氨基转移酶产生菌株的筛选鉴定

摘    要:以赖氨酸为唯一氮源,从土壤中分离到36株能够利用赖氨酸的菌株,从中筛选到2株具有赖氨酸6-氨基转移酶活力的菌株。并对酶活较高的菌株进行形态以及ITS序列鉴定,菌株2-14鉴定为轮枝霉菌(Ver-ticilliumsp.)。该菌株能将L-赖氨酸代谢转化为L-哌可酸,并且其中几乎不含有D-哌可酸。该研究为国内L-哌可酸生物合成提供了有价值的资料。

Abstract:36 strains of microorganisms were isolated from the soil sample based on lysine as sole nitrogen source and two of them were confirmed to contain the Lysine 6-aminotransferase(LAT) through the enzyme activity determination.The strain which has the higher LAT activity was verified as Verticillium sp.by the morphological and ITS sequence analysis.That the lysine metabolite of this strain was L-pipecolic acid and contained almost D-pipecolic acid by HPLC and UV absorption analysis was also found in this research.The preliminary study of Verticillium sp.2-14 shows that the strain has the ability to microbially transform L-pipecolic acid from L-lysine facilely.This study and further research of Verticillium sp.2-14 about L-lysine metabolism may lay a foundation and provide information for the domestic L-pipecolic acid biosynthesis.
