荧光激活细胞分选分别含有高比例G0/G1期、S期、G2/M期细胞亚群的研究

摘    要:目的:建立利用荧光激活细胞分选技术同时分选、获得含不同细胞周期细胞群的方法。方法:应用流式细胞仪的光学参数-前向角和侧向角,在细胞群中设置12个门,然后进行分选,分选出的各门细胞再经碘化丙啶(PI)染色进行细胞周期检测。结果:低侧向角、中前向角的细胞群内G0/G1期细胞比例可达70.0%,中侧向角、低前向角的细胞群内S期细胞比例可达72.3%,高侧向角、高前向角的细胞群内G2/M期细胞比例可达42.8%。结论:应用流式细胞仪的光学参数可以有效同时分选、获得分别含有高比例G0/G1期、S期、G2/M期的细胞亚群。

Abstract:Objective:In this study,we isolated cells in different phases of the cell cycle by FACS(flow activated cell sorting,FACS) at the same time.Method: The cells were divided and sorted to the 12 sub-fractions according to two light scatter parameters(the area of forward scatter and side scatter,FSC and SSC).The cell cycle of each sub- fraction was checked by FACS and PI staining.Results:The percent of cells in G_0/G_1 phase was 70.0% in the sub-fraction of low SSC-area and medium FSC-area.The percent of cells in S phase was 72.3% in the sub-fraction of medium SSC-area and low FSC-area.The percent of cells in G_2/M phase was 42.8% in the sub-fraction of high SSC-area and high FSC-area.Conclusion:Sorting of cell sub-fractions by the light parameters of FACS is an effective method for preparing cells containing the high percent of G_0/G_1 phase, S phase,G_2/M phase cells respectively.
