高尔基体糖蛋白73在肝癌中的表达及其影响因素分析

目的观察高尔基体糖蛋白73（Golgi protein 73,GP73）在肝细胞癌中的表达水平,分析影响其表达的相关因素。方法选取肝细胞癌、肝硬化、乙型肝炎患者及正常体检人群为研究对象,分别纳入肝细胞癌组（n=67）、肝硬化组（n=20）、乙型肝炎组（n=20）和正常对照组（n=20）。收集各组血清标本并用ELISA法检测GP73的表达水平,比较肝细胞癌组GP73表达水平与其他三组的差异。以GP73表达中位数将肝细胞癌组分为高表达和低表达亚组,分析影响GP73表达水平的相关因素。结果肝细胞癌组GP73表达最低13.8 ng/ml,最高1 128.2 ng/ml,中位表达173.1 ng/ml。将肝细胞癌组GP73表达水平分别与另外三组进行比较,肝细胞癌组GP73表达高于肝硬化组、HBV肝炎组或正常对照组（P〈0.05）。根据GP73表达中位数将肝细胞癌患者分为低于173.1 ng/ml（n=33）、超过173.1 ng/ml（n=34）两个亚组,两组患者在是否合并肝硬化、肿瘤分化程度方面差异具有统计学意义（P〈0.01）,而在年龄、TNM分期、肿瘤大小、AFP表达水平、远处转移方面差异无统计学意义（P〉0.05）。结论肝细胞癌患者血清GP73水平显著高于正常人群以及乙型肝炎、肝硬化患者;GP73表达水平与肿瘤分化程度、肝硬化有关。

Expression of Golgi protein 73 in hepatocellular carcinoma and its correlated factors analysis.

Objective To investigate the expression of Golgi protein 73 （GP73） and explore its correlated factors in hepatocellular carcinoma （HCC）. Methods The peripheral serum samples from 127 individuals were col- lected, including 67 HCC patients （HCC group）, 20 liver cirrhotic patients （cirrhotic group）, 20 hepatitis B patients （hepatitis B group） and 20 healthy people （the control group）. The expression levels of serum GP73 in hepatocellular car-cinoma were detected by ELISA, and compared with those of other groups. Using median level of GP73 in HCC group as boundary, the HCC group was divided into high expression subgroup and low expression subgroup. The correlated factors of GP73 expression were analyzed. Results The lowest expression of GP73 in HCC group was 13.8 ng/ml, while the highest was 1 128.2 ng/ml, with the median level of 173.1 ng/ml. The expression level of serum GP73 from HCC group was significantly higher than those from other groups （P〈0.05）. Based on median level, HCC group was divided into high expression subgroup （n=34, 〉173.1 ng,/ml） and low expression subgroup （n=33, 〈173.1 ng/ml）. There were significant differences in aspects of cirrhotic, tumor differentiation between the two subgroups （P〈0.01）. Conclusion GP73 expression is increased in HCC patients compared to cirrhotic patients, hepatitis B patients and healthy people. Its expression is positively correlated with cirrhotic and tumor differentiation.
