摘    要:目的:研究渥曼青霉素(Wortmannin,WT)对人恶性胶质瘤U251细胞的辐射增敏作用。方法:采用集落形成实验、流式细胞术、核增殖性抗原(proliferating cell nuclear antigen,PCNA)免疫荧光染色,以及透射电镜技术检测WT联合X线对U251细胞克隆形成及凋亡的影响。结果:WT预处理后的U251细胞接受X线照射后,集落形成率和PCNA的表达明显低于仅接受射线照射组,并且凋亡和坏死细胞数目明显增多,在透射电镜下,可见典型的凋亡和坏死细胞的形态。结论:WT作用后,明显增强了X线对U251细胞增殖的抑制作用,促进了X线引起的U251细胞凋亡和坏死的发生,增加了U251细胞的辐射敏感性。

Abstract:Objective:To study the radiative sensitization of wortmannin(WT) on human malignant glioma U251 cells. Methods:The activity of colony- forming and apoptosis of U251 cells, when treated with WT combined with X - irradiation were detected by colony - forming assay, flow cytometry, proliferating cell nuclear antigen ( PCNA) immunofluorescence staining and transmission electron microscopy. Results:The colony -forming rate and PCNA expression of U251 cells treated with WT and X - irradiation were obviously lower than that of only exposed to X - irradiation. Moreover,the numbers of apoptosis and necrosis of U251 cells treated with WT and X - irradiation were increased, and the typical morphological changes of apoptosis and necrosis were observsed in the transmission electron microscopy. Conclusion:WT increased the inhibitory effect of X -irradiation on the proliferation of U251 cells and promoted the happening of apoptosis and necrosis of U251 cells exposed to X -irradiation, WT enhanced the radiation sensitivity of U251 cells.
