缺锌对胸腺和脾脏细胞增殖周期DNA及蛋白质含量的影响

摘    要:通过建立大鼠缺锌(ZD)模型,采用流式细胞术,研究缺锌对胸腺、脾脏细胞增殖周期、DNA及蛋白质含量的影响。结果表明:(1)ZD组的最终体重、体重增长值、饲料效价、以及血清、毛发、股骨锌均非常显著地低于缺锌对喂(PF)组。(2)ZD组胸腺、脾细胞的增殖指数和G_2+M期细胞的百分率非常显著地低于PF组;胸腺、脾脏的G0/G_1期细胞百分率却非常显著地高于PF组。(3)ZD组胸腺、脾脏细胞的DNA含量、DNA指数、蛋白质含量、蛋白质指数显著低于PF组。缺锌时,DNA复制和蛋白质合成受到抑制,致使细胞停滞于G0/G_1期,仅少量进入S期和G_2+M期、使增殖指数减低。

Abstract:nalysis of the cell cycle,DNA and protein contents were performed by fluw cytometry on thymus and spleen monunuclear cell from the rats fed on zinc deficient diet(ZD) or pair-fed zinc normal diet(PF).The findings were as follows:(1)The body、weight, weight gain, feed efficiency value and the zinc content in serum,hair and femur of ZD group were significantly lower than those of PF group.(2)The proliferative index and the proportion of G_2+ M phase cells of thymus and spleen tvere significantly lower in ZD group than in PF group,but G_0/G1 phase cells were significantly higher in ZD group than in PF group.(3) The DNA content,DNA index,protein coutent and protein index of thymic and splenic cell were significantly lower in ZD group than in PF group. It was concluded that zinc deficiency could depress the syntheses of DNA and protein in cells.Numerous cells stagnated at G_0/G_1 phase,that affected the proliferation of the cells.
