| 摘 要： | seladin-1又叫做DHCR24,是催化链甾醇合成胆固醇的胆固醇合成酶,在阿尔茨海默病选择性受累的脑区表达减少。在阿尔茨海默病中seladin-1具有减少Aβ生成、对抗Aβ毒性、抑制细胞凋亡等多种保护作用。seladin-1基因表达受雌激素、胰岛素样生长因子1、雄激素、甲状腺激素和肝X受体的调节。明确seladin-1在阿尔茨海默病中的保护作用及其作用机制对于完善阿尔茨海默病发病机制,制订新的治疗策略具有重要意义。本文就seladin-1在阿尔茨海默病中的保护作用及其机制进行综述。

| Abstract: | The cholesterol-synthesizing enzyme seladin-1, also known as DHCR24, is a 3β-hydroxysteroid-Δ24 reductase responsible for catalyzing the synthesis of cholesterol from desmosterol. Its expression is downregulated in vulnerable brain regions in Alzheimer disease （AD）. Seladin-1 has various biological effects against AD, including decreasing Aβ production, conferring resistance to Aβtoxicity and inhibiting apoptosis. The expression of seladin-1 gene is regulated by estrogen, insulin-like growth factor 1, androgen, thyroid hormone and liver X receptor. This review summarized the neuroprotective roles and its underlying molecular mechanisms of seladin-1 in AD, aiming to better understand the pathogenesis of AD and offer a plausible strategy for its prevention and treatment. |
