Skp2,p27,PTEN在卵巢上皮性肿瘤中的表达和意义

摘    要:目的探讨卵巢上皮性肿瘤中Skp2,p27和PTEN表达的意义以及表达的相关性。方法用免疫组化法分别检测卵巢腺瘤(n=20),交界性卵巢腺瘤(n=20),卵巢腺癌(n=50)标本Skp2,p27和PTEN的表达。以病理分级,临床分期,和淋巴结转移作为卵巢腺癌的独立因素进行分析。结果卵巢腺癌Skp2表达率与卵巢腺瘤和交界性卵巢腺瘤相比显著增高(P〈0.01)。卵巢腺癌p27,PTEN表达率与卵巢腺瘤和交界性卵巢腺瘤相比显著降低(P〈0.01,P〈0.05)。随卵巢癌病理分级的提高,Skp2的表达显著增高,p27和PTEN的表达显著降低。随卵巢癌临床分期的进展,Skp2的表达显著增高,p27和PTEN的表达显著降低。与无淋巴结转移者相比,有淋巴结转移者Skp2表达率较高,而p27和PTEN的表达率较低。结论在卵巢腺癌中PTEN的缺失突变导致Skp2的表达升高,进而促进p27的泛素化降解,使p27表达降低,失去对细胞周期的调控,从而促进卵巢腺癌的发生发展。Skp2高表达,p27和PTEN低表达分别在人卵巢上皮性肿瘤的发生,发展中可能起一定的作用,并与预后相关。

Abstract:Objective To investigate significance of Skp2,p27 and PTEN expression in human epithelial ovarian tumor.And the relationship among Skp2,p27 and PTEN was also studied.Methods The expression of skp2,p27,PTNE were detected on resected specimens from patients suffering from adenoma(n=20),borderline adenoma(n=20),and adenocarcinoma(n=50)in Yijishan Hospital from 1999 to 2006 by Immunohistochemical analysis,In analyzing the significance of skp2,p27 and PTEN in adenocarcinoma,clinical information such as Pathologic grade,Clinical stage,and Lymph node metastasis were also evaluated as independent factors.Results Skp2 labeling frequency was increased dramatically in adenocarcinoma compared with adenoma and borderline adenoma (P<0.01).While p27 and PTEN labeling frequency was significantly lower than that of adenoma and borderline adenoma(P<0.01,P<0.05).In adenocarcinoma,with the increase of Pathologic grade,Skp2 labeling frequency increase gradually,while p27 and PTEN labeling frequency decrease gradually;with the development of clinical stage,Skp2 labeling frequency increase gradually,while p27 and PTEN labeling frequency decrease gradually.Specimens with Lymph node metastasis show a higher expression of skp2(P<0.05)and a lower expression of p27 and PTEN(P<0.05).Conclusion The expression of Skp2 was increased due to the mutation of PTEN in adenocarcinoma.Therefore the degradation of p27 in the ubiquitinpmteasome system was promoted.Skp2 may play an important role in the development of adenocarcinoma by its promotive effect on decomposition of p27 and it may function as a critical component in the pathway of PTEN regulating p27 and cell proliferation in adenocarcinoma.Skp2 overexpression,p27 and PTEN low expression may play an important role in the genesis and development of human epithelial ovarian tumor respectively and are related to the prognosis of adenocarcinoma.
