| 摘 要： | 目的探讨人工合成的含CpG基序的寡脱氧核苷酸(ODN)作为佐剂,对乙型肝炎表面抗原(HBsAg)诱导普通小鼠产生体液免疫应答的影响.方法以血源HBsAg为接种疫苗,人工合成含CpG基序的硫代磷酸寡脱氧核苷酸作为佐剂,以BALB/c小鼠作为实验动物进行免疫接种;采用ELISA方法检测抗HBs,判断免疫小鼠的体液免疫功能.结果联合CpG ODN组在抗HBs转化率、出现时间、滴度峰值方面优于单用HBsAg组,CpG ODN的佐剂效果接近完全弗氏佐剂,而单用CpG ODN则不能诱导抗HBs的产生.结论CpG ODN作为佐剂可增强HBsAg诱导BALB/C小鼠产生体液免疫应答.

| Abstract: | Objective To investigate the enhancing effect of oligodeoxyribonucleotide (ODN) containing CpG on hu- moral immune response induced by HBsAg in healthy mice. Methods Phosphorothioate oligodeoxynucleotide containing 2 CpG motifs was synthesized as the adjuvant of HBsAg. Then BALB/c mice were inoculated with the adjuvant to determine their immune response to HBsAg. Anti - HBs was detected by ELISA. Results The mice receiving HBsAg and ODN contai- ning CpG showed higher specific humoral immune responses to HBsAg than those receiving HBsAg alone. Conclusions ODN containing CpG can act as an adjuvant to enhance humoral immune responses in mice immunized with HBsAg. |
