针刺对围绝经期大鼠卵巢颗粒细胞Fas、Bcl-2 mRNA表达的影响

摘    要:目的从细胞凋亡角度研究针刺治疗围绝经期综合征的作用机制。方法将围绝经期大鼠随机分为围绝经期组、针刺组和药物组,并设青年对照组。围绝经期组大鼠不治疗,作为空白对照;针刺组大鼠针刺肾俞、足三里、三阴交治疗;药物组大鼠采用更年安药物治疗;青年对照组大鼠不治疗。应用原位杂交法检测卵巢颗粒细胞Fas、Bcl-2 mRNA表达,并进行图像分析。结果与青年对照组比较,围绝经期组大鼠卵巢颗粒细胞Fas mRNA表达明显升高(P<0.01),Bcl-2 mRNA表达明显降低(P<0.01);针刺治疗后围绝经期大鼠卵巢颗粒细胞Fas mRNA表达明显降低(P<0.01),Bcl-2 mRNA表达明显提高(P<0.01)。结论针刺能够下调围绝经期大鼠卵巢颗粒细胞Fas mRNA表达,上调Bcl-2 mRNA表达。

Abstract:Objective To investigate the mechanism of therapeutic action of acupuncture on periclimacteric syndrome from cellular apoptosis.Methods Periclimacteric rats were randomly allocated to periclimacteric,acupuncture and medication groups.A group of young rats was established as a control.The periclimacteric group of rats was not treated as a blank control.The acupuncture group of rats was treated by needling Shenshu,Zusanli and Sanyinjiao.The medication group of rats was treated with Gengnianan.The control group of young rats was not treated.The expressions of Fas and Bcl-2 mRNAs in ovarian granulosa cells were examined by in situ hybridization.Results The expression of Fas mRNA in ovarian granulosa cells significantly increased(P<0.01) and the expression of Bcl-2 mRNA significantly decreased(P<0.01) in the periclimacteric group of rats as compared with the control group of young rats.The expression of Fas mRNA in ovarian granulosa cells significantly decreased(P<0.01) and the expression of Bcl-2 mRNA significantly increased(P<0.01) in periclimacteric rats after treatment with acupuncture or Gengnianan.Conclusion Acupuncture can down-regulate the expression of Fas mRNA and up-regulate the Bcl-2 mRNA in ovarian granulosa cells in periclimacteric rats.
