缺氧缺血性脑损伤新生鼠尿8-异前列腺素F_(2a)变化及N-乙酰半胱氨酸的干预效果

摘    要:目的:探讨实验性缺氧缺血性脑损伤大鼠尿与脑组织8-iso-PGF2a相关性,尿8-iso-PGF2a能否作为监测早期脂质过氧化作用的生化指标及NAC的治疗效果。方法:7d龄SD新生大鼠60只随机分为对照组,缺氧缺血组和NAC组。NAC组给予NAC 0.1mg.g-1.d-1,于缺氧缺血前3d灌胃,共3d,缺氧缺血组给予等体积生理盐水,对照组不做相应处理。应用ELISA法测定各组大鼠尿与脑组织8-iso-PGF2a含量。结果:缺氧缺血组尿和脑组织含量分别为(101.5±22.8)ng.mmol-1.cr-1和(237.2±11.3)ng/g,明显高于对照组(26.5±11.6)ng.mmol-1.cr-1和(90.8±18.7)ng/g(P<0.01),NAC组(40.1±9.9)ng.mmol-1.cr-1和(110.5±12.4)ng/g,明显低于缺血组(P<0.01),尿与脑组织8-iso-PGF2a水平相关(r=0.823,P<0.01)。结论:急性缺氧缺血性脑损伤时尿8-iso-PGF2a含量增加,NAC能有效降低尿8-iso-PGF2a水平,可用于治疗缺氧缺血性脑损伤。

Abstract:Objective:To analyze the correlation of 8-iso-prostaglandin F2a levels in both urine and brain tissue of newborn rats with cerebral hypoxia-ischemia,and to investigate the rapeutic effect of NAC,and the possibility of level of 8-iso-PGF2a in urine as a kind of biochemical index.Methods: 60 Seven-day-old Spraygue-Dawley(SD) rats were randomly divided into 3 groups:hypoxia-ischemia,NAC and control group.Rats in the NAC group were given gastric lavage with NAC(0.1mg/g,perday) for three days.Rats in hypoxia-ischemia group were given an equal volume of normal saline,and control group had no administration.The 8-iso-PGF2a contents in urine and brain tissue were determined by ELISA.Results:Compared with the control group.8-iso-PGF2a contents in urine and brain-tissue of the hypoxia-ischemia group were significantly higher(26.5±11.6)ng·mmol-1·cr-1 vs(101.5±22.8)ng·mmol-1·cr-1 and(90.8±18.7)ng/g vs(237.2±11.3)ng/g(P<0.01 for all).In the NAC group,8-iso-PGF2a concentration of urine and brain tissue were(40.1±9.9)ng·mmol-1·cr-1 and(110.5±12.4)ng/g,respectively,lower than those in the hypoxia-ischemia group(P<0.01).The 8-iso-PGF2a levels of urine and brain tissue were positively correlated(r=0.823,P<0.01).Conclusion:The levels of 8-iso-PGF2a increases in both urine and brain tissue of rats with acute cerebral hypoxia-ischemia.NAC can reduce the level of 8-iso-PGF2a in urine.Therefore it may be used in treatment of cerebral hypoxia-ischemia.
