不同浓度丙泊酚对大鼠单肺通气时肺损伤因子的影响

摘    要:目的观察不同浓度丙泊酚对大鼠单肺通气时急性肺损伤的保护作用,为丙泊酚在胸科麻醉中的应用提供实验研究的数据。方法 40只健康清洁级SD大鼠随机分成四组:P1组:空白组(灌注液中无丙泊酚)、P2组:含36mg/L丙泊酚、P3组:含24mg/L丙泊酚、P4组:含12mg/L丙泊酚,每组10只。按随机顺序取大鼠进行实验,称量体重,麻醉,气管切开插管接呼吸机行单肺机械通气1h。然后取出肺组织。取双侧顶叶头盖型部分作湿/干比,其余-70℃保存以备统一作髓过氧化物酶(MPO)以及微量丙二醛(MDA)测定。结果肺组织湿/干重比:P1组通气侧高于P2、P3组通气侧(P<0.05);P1、P4组非通气侧高于P2、P3组非通气侧(P<0.05)。各组非通气侧均高于同组通气侧(P<0.01)。肺组织丙二醛(MDA)含量:P1组通气侧肺较P2、P3和P4组通气侧高(P<0.05);P1组非通气侧较P2、P3和P4组非通气侧显著增高(P<0.01)。各组非通气侧均显著高于同组通气侧(P<0.01)。肺组织髓过氧化物酶(MPO)含量:P1组通气侧高于P2、P3、P4组通气侧(p<0.05),P4组通气侧高于P2、P3组通气侧(P<0.05);P1组非通气侧高于P2、P3、P4组非通气侧。各组非通气侧均高于同组通气侧(P<0.01)。结论通过对大鼠单肺通气时的灌注实验研究,发现丙泊酚对单肺通气时的急性肺损伤有保护作用,其强度在一定范围内随着丙泊酚浓度增加而增强。

The effect of propofol of different concentrations on injury factor of rats lung during one-lung ventilation

Abstract:【Objective】To investigate the effect of propofol of different concentrations on injury factor vasoconstriction of rats lung during one-lung ventilation.【Methods】 The effects of 0,36,24and12mg/L propofol on injury factors were assessed in isolated lungs of rat.40rats randomly divided into 4 groups(n=10,each): group P1(no propofol in the perfusate),group P2(the concentration of propofol in perfusate was 36mg/L),group P3(24mg/L),group P4(12mg/L).Injury factors was quantified after one-lung ventilation.After OLV finished,lung tissue sample were harvest for the ratio of wet to dry weight(WDR),the ration of MDA and the ration of MPO.【Results】 1)The ratio of wet to dry weight(WDR): as compared to the values in group P1 and P4 ventilated lungs,the WDR was low in group P2 ventilated lung and group P3 ventilated lung(P<0.05);as compared to the values in group P1 airproof lung and group P4 airproof lung,the WDR was low in group P2 airproof and group P3airproof(P<0.05).as compared to the values in reciprocal group airproof lung,the WDR was low in each group ventilated lung(P<0.01).2)The ratio of MDA: as compared to the values in group P1 ventilated lung,the ration of MDA was low in group P2 、group P3 and P4 ventilated lung(P<0.05);as compared to the values in group P1 airproof lung,the ration of MDA was significantly low in group P2 、group P3 and group P4 airproof lung(P<0.01).as compared to the values in group P2 and group P4 airproof lung,the ration of MDA was significantly low in group P3 airproof lung(P<0.05);as compared to the values in reciprocal group airproof lung,the ration of MDA was low in each group ventilated lung(P<0.01).3)The ratio of MPO: as compared to the values in group P1 ventilated lung,the ration of MPO was low in group P2 、 group P3 and group P4ventilated lung(P<0.05),as compared to the values in group P4 ventilated lung,the ration of MPO was low in group P2 and group P3 ventilated lung(P<0.05);as compared to the values in group P2 、 group P3 and group P4airproof lung the ratio of MPO was high in group P1 airproof lung(P<0.01),as compared to the values in group P3 airproof lung,the ratio of MPO was high in group P4;as compared to the values in reciprocal group airproof lung,the ratio of MPO was low in each group ventilated lung(P<0.01or0.05).【Conclusion】 These results indicate that the propofol inhibits lung injury factor of rats during one-lung ventilation,and the inhibition increase as the concentration of propofol increase,however,the increase can stop as the concentration reaches peak value.
