西红花酸对压力超负荷致心肌肥厚大鼠的ATPase及胶原的影响

摘    要:目的 研究西红花酸对压力超负荷所致心肌肥厚大鼠的ATPase及胶原含量的影响。方法 采用腹主动脉部分狭窄术致大鼠心肌肥厚模型,大鼠分为早期组和晚期组,又各自分为假手术对照组、模型组、卡托普利组、西红花酸100,50mg/kg组。测定心脏重量参数,采用试剂盒测定Na^ ,K^ —ATPase;Ca^2 ,Mg^2 —ATPase的活力及羟脯氨酸的含量。结果 模型早期组(术后24h)ATPase活性明显降低,模型晚期组(术后1个月)ATPase活性降低更加明显,羟脯氨酸的含量明显增加。西红花酸晚期组(给药1个月)能显著提高心肌组织的ATPase活力,降低胶原的含量。结论 西红花酸对压力超负荷所致大鼠心肌肥厚具有一定的改善作用。

Abstract:Object To study the influence of crocetin on the activity of ATPase and the content of hydroxyproline in the myocardial hypertrophy rats induced by overload pressure. Methods Abdominal aortic ligation was used to establish the myocardial hypertrophy model in rats. The rats were divided into two groups: 1) The early term group which included five groups: sham-operation group, model group, Captopril groups, crocetin 100, 50 mg/kg groups. 2) The long term group which also included five groups as the same as the early term groups. Heart indexes were examined. The ATPase activity of heart and the content of hydroxyproline in heart were assayed by colorimetric analysis. Results Compared with the model group, crocetin can significantly reduce the cardiac indexes, increase the activities of Na +, K +-ATPase; Ca 2+ , Mg 2+ -ATPase and markedly decrease the content of hydroxyproline in heart. Conclusion Crocetin could prevent the remodeling of cardiac hypertrophy induced by overload pressure. The ATPase may play an important role in myocardial hypertrophy induced by overload pressure.
