miR-101在维吾尔族妇女子宫颈癌中的表达及其与COX-2的关系

摘    要:目的研究miR-101与其靶基因环氧合酶-2(Cyclooxygenase,COX-2)在维吾尔族妇女慢性宫颈炎、宫颈上皮内瘤样病变Ⅰ、Ⅱ、Ⅲ(CINⅠ、Ⅱ、Ⅲ)及子宫颈癌组织中的表达,以探讨miR-101及其与COX-2的关系。方法应用锁定核苷酸原位杂交技术(LNA-ISH)检测维吾尔族妇女慢性宫颈炎、CINⅠ~Ⅲ和子宫颈癌组织中miR-101的表达情况;应用免疫组化Envision两步法检测COX-2在维吾尔族妇女慢性宫颈炎,宫颈上皮内瘤样病变Ⅰ、Ⅱ、Ⅲ及子宫颈癌组织中的表达情况,并进行相关性分析。结果 miR-101在慢性宫颈炎、CINⅠ~Ⅲ及子宫颈癌中的表达率分别为80%、72%、48%、24%和8%,差异具有统计学意义(χ2=37.828,P=0.000)。慢性宫颈炎组织中未见COX-2阳性表达;CINⅠ、Ⅱ、Ⅲ以及子宫颈癌组织中COX-2阳性表达率分别为12%、36%、60%、84%,差异具有统计学意义(χ2=28.846,P<0.05)。miR-101与其靶基因COX-2呈负相关性(R=-0.997,P=0.000)。结论 miR-101与其靶基因COX-2可能与子宫颈癌的发病机制有关。

Abstract:Objective To detect the expression of miR-101 and the target genes COX-2 in cervical intraepithelial neoplasiab Ⅰ,Ⅱ,Ⅲ(CINⅠ,Ⅱ,Ⅲ) and cervical cancer in Uighur women.Methods We investigated the expression profiles of miRNA-101 using the method of locked nucleic acid(LNA)-modified oligonucleotide in Situ Hybridization(ISH) with the formalin-fixed paraffin-embedded(FFPE) specimens from chronic cervicitis,CINⅠ,Ⅱ,Ⅲ and cervical cancer.The expressions of COX-2 in chronic cervicitis,CINⅠ,Ⅱ,Ⅲ and cervical cancer tissue specimens obtained by immunohistochemical staining.Then we analyzed the correlation between the expressions of miR-101 and the target genes COX-2 in cervical cancer.Results The positive rate of miR-101 in patients with chronic cervicitis,CINⅠ~Ⅲ and expression in cervical cancer rates were 80%,72%,48%,24% and 8%,a statistically significant difference(χ2=37.828,P=0.000).No positive expression of COX-2;chronic cervicitis tissues of CIN Ⅰ,Ⅱ,Ⅲ and cervical cancer,COX-2 positive expression rates were 12%,36%,60%,84%,a statistically significant difference(χ2=28.846,P<0.05).miR-101 and its target gene COX-2 was a negative correlation(R=-0.997,P=0.000).Conclusion miR-101 and the target genes COX-2 involved in development and progresstion of cervical cancer.
