大鼠闭合性脑损伤后5-LOX、c-fos蛋白、NSE的表达及表达时相的研究

摘    要:目的探讨5-脂氧合酶(5-LOX)、c-fos蛋白及神经元特异性烯醇化酶(NSE)在闭合性脑损伤后不同时间点的表达情况及其意义。方法 Wistar大鼠55只随机分成正常对照组、假损伤组及损伤后0.5h组、1h组、2h组、4h组、8h组、12h组、24h组、48h组、72h组。用免疫组化(SP)法检测5-LOX、c-fos蛋白和ELISA检测NSE在闭合性脑损伤后不同时间点的表达情况。结果 5-LOX免疫阳性细胞在损伤后8h明显增高,24h达到高峰,然后逐渐降低,损伤72h后仍高于正常值;c-fos蛋白于损伤0.5h后表达明显增高,8h达到高峰,24h降至正常值;血清中的NSE于损伤2h候表达增高,24小时达到高峰,损伤72h后仍高于正常值。结论大鼠闭合性脑损伤后5-LOX、c-fos蛋白及血清中的NSE表达均上调,且各自具有时间规律,有希望为法医学中的颅脑损伤时间的推断提供参考。

Study on expression and its sequence of 5-LOX,c-fos albumen,and NSE after closed traumatic brain injury in rats

Abstract:Objective To study the time-dependent expression features and the significances of 5-LOX,c-fox albumen,neuron-specific enolase(NSE) in rats with closed traumatic brain injury(TBI).Methods Fifty Wistar rats were randomly dividied into control group,sham group and 9 time-dependent experimental groups which were tested in 9 different time such as 0.5,1,2,4,8,12,24,48 and 72 h.The expression and its sequence of 5-LOX,c-fos albumen and NSE were assessed through the immuohistochemisty SP method.Results There was an obvious increasing after immunoreactive positive cells of 5-LOX had been injured in 8 h,and it reached the highest point in the 24 th hour,then it went down gradually.However,it was still higher than the normal level after 72 h.It was different that the ratio of the c-fos albumen rose rapidly after injured,and it reached the peak level in the 8 h,then decreasing to the normal level in the 24 h.With the comparison of the above,the ratio of NSE in the serum went up clearly,and reached to the top in the 24 h.It was similar with the 5-LOX;NSE was still higher than normal after 72 h.Conclusion Closed traumatic brain injury induced expression of 5-LOX,c-fos,and NSE.The time-dependent expression feature might provide evidence for brain injury in forensic detection.
