氟对大鼠水迷宫学习记忆及脑内单胺类神经递质浓度的影响

摘    要:目的探讨氟对大鼠学习记忆及脑内单胺类神经递质浓度的影响。方法 40只SD大鼠随机分为对照组及低、中、高剂量染毒组,分别以0、5、10、20mg/kg的氟化钠(NaF)对SD大鼠染毒90d后,应用水迷宫测试其学习记忆能力,并采用ELISA法检测脑组织中多巴胺(DA)和5-羟色胺(5-HT)浓度。结果高剂量染毒组与对照组大鼠水迷宫实验中抵达终点的游泳时间差异有统计学意义(P<0.05),其它各组抵达终点的游泳时间差异均无统计学意义(P>0.05);与组对比,高剂量染毒组脑中DA含量明显下降,差别有统计学意义(P<0.05);其它各剂量组DA及5-HT含量与对照组相比,差异均无统计学意义(P>0.05)。结论氟对大鼠学习记忆的影响可能与脑内单胺类神经递质含量变化有关。

Abstract:Objective To study the effects of fluoride exposure on performance in water labyrinth and monoamine neurotransmitters of rats.Methods Fourty SD rats were divided randomly into 4 groups:control group,high-fluoride exposed group,medium-fluoride exposed group and low fluoride exposed group.These 4 groups of rats were exposed to different concentrations(0,5,10,20 mg/kg) of sodium fluoride(NaF) for 90 consecutive days,and their behaviors that they learned to perform a water labyrinth task and monoamine neurotransmitters(DA and 5-HT) levels were tested.Results There were no significant difference in swimming duration among 0,5 and 10 mg/kg groups,while rats in 20 mg/kg group had notable more swimming time(P<0.05),and DA level significantly decreased compared with the control group(P<0.05),no significantly difference in 5-HT level were found among all groups.Conclusion The results demonstrate that fluoride exposure may develop an adverse effect on the learning capacity of rats,this may be caused by monoamine neurotransmitters levels alteration.
