摘    要:目的考察新疆乌头属植物总生物碱含量测定的方法。方法用分光光度法测定新疆乌头属植物中总生物碱的含量。以乌头碱为对照品,溴甲酚绿为酸性染料,缓冲溶液pH(3.0±0.1)条件下,三氯甲烷萃取45min,于415nm波长处测定其吸光度。结果乌头碱在11.76~27.44mg/L浓度范围内线性关系良好,回归方程Y=21.403 X+0.003 5,r=0.999 1,精密度良好,RSD=0.99%;样品溶液显色后4h内稳定,RSD=1.27%。本法测定的重复性良好,RSD=1.37%;平均加样回收率为100.89%,RSD=1.82%。新疆伊犁不同地区分布的白喉乌头和准噶尔乌头较多,其总生物碱含量平均值分别为0.236%和0.238%。结论分光光度法简便、准确,可用于新疆乌头属植物中总生物碱的含量测定。

Abstract:Objective To investigate the method the determination of the content of total alkaloid in Xinjiang genus Aconitum.Methods Using spectrophotometry to determine the content of total alkaloid in Xinjiang genus aconitum.Using aconitine as reference,bromocresol green as acid dye,buffer solution pH=3 conditions,chloroform extraction 45 min,measured at 415 nm wavelength absorbance.Results Aconitine in 11.76-27.44 mg/L concentration range a good linear relationship,the regression equation Y=21.403 X+0.003 5,r=0.999 1 good precision,RSD=0.99%;color sample solution was stable after 4 h,RSD=1.27%.Applied to the determination of the good reproducibility,RSD=1.37%;average recovery was 100.89%,RSD=1.82%.Conclusion The method is simple,accurate,and can be used for the determination of total alkaloids in the genus aconitum of Xinjiang.
